甲基强的松龙和脑源性神经营养因子联合使用对鼠脊髓损伤的治疗作用

摘    要:目的:观察甲基强的松龙(MP)和脑源性神经营养因子(BDNF)联合治疗对鼠脊髓损伤后结构修复和功能恢复的作用,探讨一种有效的急性脊髓损伤的治疗方案。方法:应用微型恒量灌注泵硬膜下灌注BDNF和静脉注射MP的联合治疗方法,利用鼠T10脊髓损伤模型观察MP和BDNF联合治疗对脊髓损伤后结构修复和功能恢复的影响,结果:与对照组相比,MP和BDNF联合治疗明显减轻了炎症细胞的浸润和继发性损伤,促进了脊髓运动神经元轴索的再生和下肢运动功能的恢复(P<0.05),结论:MP和BDNF联合治疗对脊髓损伤后结构修复和功能恢复有明显的促进作用,是急性脊髓损伤一种有效的联合治疗方案。

Abstract:Objective To investigate the effects of combined therapy of methylprednisolone (MP) and brain derived neurotrophic factor (BDNF) on morphological repairing and functional recovery after spinal cord injury in rats.Methods Contusion injury to rat spinal cord was produced at T 10 vertebral level and received intravenous MP immediately,and followed by intrathecal infusion of BDNF for 10 weeks.Results Combined therapy of MP and BDNF resulted in a decrease in inflammatory cellular infiltration,an increase in motor axonal regeneration and functional recovery significantly as compared to the controls (P<0.05).Conclusion Combined therapy of MP and BDNF could improve functional recovery by way of mechanisms that include attenuating inflammatory cellular infiltration and enhancing motor axonal regeneration at the injury site.This therapy is an effective approach for treatment of spinal cord injury.
