摘    要:目的:使用VEGF shRNA真核表达载体转染肝癌细胞系SMMC-7721,观察对其增殖影响。方法:使用VEGF shRNA真核表达载体转染肝癌细胞系SMMC-7721,ELISA法检测转染前后肝癌细胞系SMMC-7721的VEGF蛋白表达量,检测转染前后肝癌细胞系SMMC-7721的增殖及细胞的克隆形成能力的变化。结果:VEGF shRNA真核表达载体大部分阻断肝癌细胞系SMMC-7721的VEGF蛋白表达,转染后细胞周期中,G1期细胞增多,S期细胞减少,细胞增殖能力降低;MTT法观察细胞生长速度变慢,单个细胞的克隆形成能力明显下降。结论:VEGF shRNA真核表达载体可明显抑制SMMC-7721细胞增殖能力,为将VEGF shRNA真核表达载体应用于肝癌的基因治疗提供依据。

Abstract:Objective:To investigate the effects of human hepatocellular carcinoma cell line SMMC-7721 transfected with eukaryotic expression vector carrying VEGF shRNA.Methods:The vector was transfected into SMMC-7721 and the expression of VEGF of SMMC-7721 before and after transfection was detected by ELISA.The biological characteristics of SMMC-7721 before and after the transfection were observed.Results:The expression of VEGF of SMMC-7721 was blocked by VEGF shRNA,and the G1 phase cells were increased and the S phase cells were decreased in cell cycle,and the clone formation ability of single cell was decreased after transfection.Conclusion:VEGF shRNA eukaryotic expression vector might have potential role in the gene therapy for hepatocellular carcinoma.
