卡介菌多糖核酸对小鼠DC2.4细胞的影响

摘    要:目的:研究卡介菌多糖核酸(BCG-PSN)对小鼠DC2.4细胞的影响.方法:用5 μg/mL(BCG-PSN5组)、20 μg/mL(BCG-PSN20组)、50 μg/mL(BCG-PSN50组)BCG-PSN刺激小鼠DC2.4细胞,用倒置显微镜观察细胞生长状态,流式细胞仪检测主要组织相容性复合物(MHC-Ⅱ)类分子及协同刺激分子CD83、CD86的表达水平,用ELISA方法检测上清液中IL-12的含量.结果:BCG-PSN20组、BCG-PSN50组的CD83、CD86、MHC-Ⅱ分子表达水平高于空白对照组(P〈0.05),IL-12的浓度亦高于空白对照组;BCG-PSN5组与空白对照组差异无统计学意义(P〉0.05).结论:BCG-PSN可能通过促进DC2.4细胞成熟和分泌IL-12调节Th1/Th2平衡,发挥治疗AD的作用.

Abstract:Objective:To elucidate the pharmacological action of BCG-PSN by investigating the effects of BCG-PSN on murine DC2.4 cells. Methods: Murine DC2.4 cells were treated with BCG-PSN in concentration of 5μg/mL (BCG-PSN5 group), 20μg/mL (BCG-PSN20 group) and 50 μg/mL (BCG-PSNso group) , respectively. Growth features of the cells was observed with inverted microscope. The expression level of MHC- Ⅱ and co-stimulating molecules CD83 and CD86 was determined by flow cytometry. IL-12 level in the supernatant was detected with ELISA. Resuits: The expression level of CD83, CD86 and MHC- Ⅱwas significantly increased in both BCG- PSN20 and BCG-PSN50 groups, compared to the untreated control (P 〈 0.05 ). IL-12 level in the culture supernatant was also higher in these two groups than in the untreated control group. No significant differences were observed on expression levels of these molecules between BCG-PSN 5 group and the untreated control group. Conclusion:BCG-PSN promotes the maturation of mouse DC2.4 cells.
