| 摘 要： | 目的对血清样品miRNA表达水平变化的实时定量PCR检测进行观察,同组织样品的miRNA的变化趋势进行比较,以评价血清miRNA在原发性肝癌早期诊断中的价值。方法通过荧光定量PCR检测miRNA-125a、-200a、-199a等3个有代表性的miRNA指标在正常和癌变样品中的变化,验证组织及血清两类样品中miRNA的变化趋势,分别取癌旁组织和癌组织以及正常人与癌症患者血清为材料。结果肝癌患者血清是正常对照血清中miRNA的含量的10%～15%,肝癌组织中的miRNA含量是癌旁组织的10%～20%,而且血清miRNA的变化趋势与组织miRNA较一致。结论血清miRNA可以反映原发性肝癌的发生,说明血清miRNA非常有可能成为一种新的生物标记物用于原发性肝癌的早期诊断。

| Abstract: | Objective The evaluate the use of miRNA as biomarker for the early diagnosis of primary hepatic carcinoma（PHC）.Methods Three representative miRNAs（miRNA-125a,-200a and-199a） were selected in this study.Their expression in serum and tissues was monitored by real-time PCR.Results The expression levels of all the three miRNA in serum from PHC patients were about 10%～15% of the normal serum.Similarly,their expression level in PHC tissues was also 10%～20% lower than their adjacent tissues.Conclusion The level of serum miRNA125a,-200a and-199a can be used as biomarkers for the early diagnosis of PHC. |
