HBV基因型与HBx基因变异的研究

目的分析慢性HBV感染者血清中HBV基因型和HBx基因多态性,探讨HBx基因变异与HBV基因型的关系。方法采用型特异性引物PCR法对110份慢性HBV感染者血清中HBV基因型进行检测。采用巢式PCR、单链构象多态性与异源双链分析、以及基因测序和生物信息学方法分析HBx基因突变。结果在110份慢性HBV感染者血清中,检出B型、C型和混合型B＋C,分别占57份（56.8%）、49份（44.6%）和4份（3.6%）。与参考序列相比,B型与C型HBx基因都存在点突变,且C型点突变数明显多于B型（t=-12.599,P〈0.05）。结论慢性HBV感染者HBx基因突变与HBV基因型密切相关。

Study on the Polymorphism of Hepatitis B Virus X Gene in Patients with Chronic HBV Infection

Objective To study the mutation of hepatitis B virus X gene （HBx） in the patients with chronic HBV infection and investigate whether there were the relationship between HBV genotype and the mutation of HBx. Methods HBV genotypes were determined by nested-PCR using type-specific primers in 110 serum samples of patients with chronic HBV infection, HBx was examined by nested PCR, agar gel electrophoresis （AGE）, single-stranded conformational polymorphism （SSCP） and heteroduplex analysis （HA）. The HBx genes with potentially mutations were cloned, sequenced and aligned with the published HBx gene sequence. Results Of 110 patients with chronic HBV infection, 57（56.8%）, 49（44.6%） and 4（3.6%） were classified as genotype B, C and mixed B＋C, respectively. Point mutations were found in the HBx gene of serum samples of patients with chronic HBV infection, and there are some difference between the HBx mutation in the genotype B and that in the genotype C （t=12.599,P〈 0.05）. Conclusion Mutations of HBx from patients with chronic HBV infection are associated with HBV genotype.
