加味镇眩颗粒对高血压大鼠心肌基质金属蛋白酶-2和微血管密度的影响

摘    要:目的观察加昧镇眩颗粒对自发性高血压大鼠(SHR)血浆血管紧张素Ⅱ(AngⅡ)、心肌基质金属蛋白酶-2(MMP-2)表达水平和微血管密度(MVD)的影响。方法将50只SHR大鼠随机分为加味镇眩颗粒高剂量组、加味镇眩颗粒中剂量组、加味镇眩颗粒低剂量组、卡托普利组、SHR对照组,经过12周的干预,分别测定动物血压变化,血浆AngⅡ含量、免疫组织化学法检测心肌MMP-2蛋白表达水平、心肌微血管密度。结果治疗组均可降低SHR血压、血浆AngⅡ含量、心肌MMP-2蛋白表达水平,增加SHR心肌微血管密度,与SHR组相比差异具有统计学意义(P〈0.05),中药组中,加味镇眩颗粒高剂量组效果最好。与卡托普利组相比,高剂量组在12周末降压效果相当(P〉0.05),且更能降低心肌MMP-2蛋白表达水平(P〈0.05),增加SHR心肌微血管密度(P〈0.05)。结论加味镇眩颗粒可降低SHR血压,可能与降低血浆血管紧张素Ⅱ含量.下调心肌MMP-2蛋白表达水平,改善SHR大鼠心肌微血管稀疏的作用有关。

Effects of Zhenxuan Granules Treatment on the Expression of Myocardial Matrix Metalloproteinase-2 and Microvessel Density in Rats with Spontaneously Hypertensivon

Abstract:Objective To examine the effect of Zhenxuan granules treatment on the level of plasma Ang II, expression of myocardial matrix metalloproteinase-2 and the microvessel density in spontaneously hypertensive rats (SHR). Methods 50 male SHRs were randomly divided into 5 groups, namely the high-dose, middle-dose and low-dose Zhenxuan granule treatment group, captopril group and the SHR control group. Blood pressure was recorded from the tail artery at the beginning and every 4 weeks during the 12 weeks treatment. At the end of the experiment, the animals were sacrificed. The level of plasma AngⅡ was measured. Immunohistochemical staining was performed to determine the expression of myocardial matrix metalloproteinase-2 and microvessel density. Results Zhenxuan Decoction was found to decrease the blood pressure, plasma AngⅡ, myocardial matrix metalloproteinase and to improve the microvessel density of SHR. Result in the high-dose treatment group was more significant than the SHR control group (P〈0.05). The effect of high-dose Zhenxuan granule was similar to the captoril group in decreasing blood pressure (P〉0.05), In contrast, high-dose Zhenxuan granule was effective in decreasing myocardial matrix metalloproteinase and improving mierovessel density of SHR (P〈0.05) at 12 weeks after the treatment. Conclusion Zhenxuan granules can decrease the blood pressure, plasma Angll and myocardial matrix metalloproteinase expression, and improve myocardial microvessel density of SHR.
