微血管密度与血管生成素2在结直肠癌中的表达及意义

摘    要:目的探讨微血管密度(MVD)和血管生成素2(Ang-2)在结直肠癌中的表达和相互关系,以及它们在血管生成中的作用;探讨CD105作为新生血管标记物的临床价值。方法运用免疫组织化学技术检测CD105,Ang-2和CD34蛋白在80例结直肠腺癌组织和40例正常肠组织中的表达情况,并计算MVD,分析其表达与临床病理因素之间的关系。结果①MVD及Ang-2在结直肠癌组织与正常肠组织间表达有差异,差异具有非常显著性意义(P<0.01,P<0.01);②以CD105和CD34测得的MVD差异具有非常显著性意义(P<0.01);③Ang-2在结直肠癌中的表达与CD105的表达呈显著正相关(P<0.01),而与CD34的表达无关(P>0.05);④结直肠癌组织中以CD105测得的MVD和Ang-2的表达与浸润深度、淋巴结转移及Dukes分期有关,差异具有非常显著性意义(P<0.01);而与性别、年龄及分化程度无关(P>0.05)。结论①CD105和Ang-2均与结直肠癌血管生成有关,二者协同在结直肠癌生长、浸润及转移过程中发挥重要作用;②CD105在结直肠癌组织新生血管中有良好表达,以CD105测得的MVD可以被认为是与肿瘤生长、转移及预后有关的一项重要指标;CD105作为结直肠癌新生血管有价值的标记物,对于结直肠癌的早期诊断、判断预后和综合治疗具有重要临床意义。

Abstract:Objective To explore the significance of expression of microvascular density(MVD) and the relationship between Ang-2,MVD and colorectal cancer(CRC),to determine their effect in angiogenesis,and to determine the probability and clinical significance of CD105 as a marker of tumor-related angiogenesis.Methods Immuno-histochemical technique was used to detect the expression of CD105,Ang-2 and CD34 proteins in 80 samples of colorectal cancer and 40 samples of normal colorectal tissue,then the amount of MVD was calculated and the relationship between its expression and clinicopathological manifestations had been analyzed.Results ①The expression of MVD and Ang-2 in CRC tissue was significantly higher than that of normal colorectal tissue(P0.01).② The expression of MVD tested by anti-CD105 in colorectal cancer and normal colorectal tissues was significantly higher than that expressed by anti-CD34(P0.01).③The expression of Ang-2 was significantly correlated with the expression of CD105(P0.01),but it was not correlated with the expression of CD34(P0.05).④The expression of MVD by anti-CD105 and the expression of Ang-2 were closely associated with depth of infiltration,lymph node metastasis and Dukes classification of stages of CRC(P0.01),and their expression was not associated with gender,age and degree of differentiation of CRC(P0.05).Conclusion ①CD105 and Ang-2 were associated with angiogenesis of CRC,they had a coordinated effect in the growth,infiltration and metastasis of CRC.②The expression of MVD by CD105 can be considered as an important index,and it is associated with tumor growth,metastasis and prognosis.CD105 used as an valuable marker of angiogenesis has important clinical significance in early diagnosis,evaluation of prognosis and comprehensive treatment of CRC.
