摘    要:目的 研究八氯二丙醚吸入染毒对小鼠外周血细胞和肝、脾、肺细胞DNA的损伤作用 ,了解其损伤的可能靶部位。方法 用八氯二丙醚对小鼠进行静式吸入染毒 ,采用微量血单细胞凝胶电泳技术(SCGE) ,检测不同染毒浓度 (2 82 ,5 6 4 ,112 8mg/m3 )及不同染毒时间 (每天 1h ,连续染毒 3,6 ,9,12 ,15d)八氯二丙醚对小鼠外周血细胞的DNA的损伤情况。结果 染毒后小鼠血细胞DNA出现了明显损伤 ,表现为第 3天时DNA损伤程度最高 ,随后第 6天、 9天、 12天、 15天损伤降低并趋于稳定。肝、肺及脾细胞DNA亦出现了明显损伤 ,且存在明显的剂量 -反应关系 (P <0 0 5 ) ,其中以肺细胞DNA损伤程度最重。结论 八氯二丙醚吸入染毒可引起小鼠外周血细胞及肝、脾、肺细胞DNA的损伤 ,提示肺细胞可能是主要靶细胞

Abstract:Objective To study effects of octachlorodipropyl ether(S 2) inhalation on DNA damage in peripheral blood cells and liver,spleen and lung cells of mice,and to understand what the main target organs are in vivo test.Methods Static inhalation was applied to expose mice to S 2,using single cell gel electrophoresis(SCGE)assay to detect DNA damages in peripheral blood cells of mice at different concentrations(282 mg/m 3,564 mg/m 3 and 1 128 mg/m 3)and at different exposure time(3d,6d,9d,12d and 15d).Results DNA of peripheral lymphocytes was significantly damaged after exposure,and the most severe damage was found at the 3 rd day,and then began to decrease at the 6 th ,9 th ,12 th and 15 th day until it tended to be stable.DNA in liver,spleen and lung cells was also significantly damaged,among which DNA in lung cells was most damaged.Moreover,good dose response relationships(P<0 05)were shown in liver and lung cells.Conclusions Inhalation of S 2 could lead to DNA damage in mice peripheral lymphocytes and also in liver,spleen and lung cells and lung cells suggest being the main target organ.
