体内和体外实验观察骨形成蛋白在骨肉瘤中的作用

目的　研究骨形成蛋白 ( bone morphogenetic protein,BMP)在骨肉瘤中的表达及其与肿瘤增殖活性的关系 ,并观察 BMP单抗体外对骨肉瘤细胞作用 ,为骨肉瘤的单抗治疗打下基础。方法　应用 BMP单抗和免疫组化方法检测骨肉瘤中 BMP蛋白的表达 ;并应用增殖细胞核抗原 ( proliferating cell nuclear antigen,PCNA)的单抗对骨肉瘤的增殖活性进行检测 ;体外实验观察 BMP单抗对骨肉瘤细胞 OS-73 2的作用。结果　 2 4例标本中有 18例有 BMP蛋白的表达 ;有 BMP表达的骨肉瘤有较高的增殖活性 ;应用 BMP单抗体外可以对骨肉瘤细胞系 OS-73 2细胞产生生长抑制。结论　 BMP有可能在肿瘤的发生和发展过程中起一定作用 ,可以作为估计骨肉瘤预后的参考指标 ,BMP单抗对骨肉瘤细胞的抑制作用为其临床应用于骨肉瘤的单抗治疗打下基础

IN VIVO AND IN VITRO STUDY OF THE EFFECT OF BMP ON OSTEOSARCOMA

Objective To study the relationship between bone morphogenetic protein(BMP) expression and proliferating activity of osteosarcoma and detect the effect of BMP antibody on osteosarcoma cell.Methods BMP and proliferating nuclear protein(PCNA) expression were examined in 24 specimens of osteosarcoma with immuno histochemistry technique. The effect of BMP antibody on osteosarcoma cell line OS 732 was studied in vitro.Results BMP positive reaction was observed in 18/24 specimens, the specimens with BMP had more positive PCNA. In vitro BMP antibody could inhibit the proliferating of osteosarcoma cell.Conclusion BMP may play some roles in the development of osteosarcoma and may be helpful markers to determine the prognosis of osteosarcoma and there may be feasibility to treat osteosarcoma with BMP antibody.
