莫诺苷抑制过氧化氢诱导的神经细胞损伤作用

摘    要:目的研究从中药山茱萸提取的有效成分莫诺苷抑制过氧化氢(H2O2)诱导的神经细胞氧化损伤作用。方法培养的人神经母细胞瘤细胞系SH-SY5Y细胞加入莫诺苷1、10、100μmol/L预孵育24h,加入H2O2300μmol/L作用18h诱导产生氧化损伤,测定细胞内细胞色素C及脑源性神经生长因子(BDNF)的含量。结果莫诺苷1、10、100μmol/L使细胞色素C的释放显著降低18%(P<0.05)、28%(P<0.05)、35%(P<0.01);BDNF(10、100mol/L)的释放增加(P<0.01)。结论莫诺苷的神经保护作用是通过抑制细胞色素C的释放,抑制神经细胞凋亡,并能促进神经细胞增加神经营养因子的释放,进而促进神经细胞的再生,抑制神经细胞氧化损伤。

Abstract:Objective To investigate the effects of morroniside on H2O2-induced oxidative injury in SH-SY5Y neuroblastoma cells,which was extracted from the sarcocarp of C.officinalis.Methods SH-SY5Y cells were pre-in-cubated with morroniside(1,10 and 100μmol/L)for 24 h prior to exposure to H2O2(300μmol/L)for 18 h.The Re-lease of cytochrome C(Cyt C)and the expression of brain-derived neurotrophic factor(BDNF)were respectively deter-mined by ELISA and Western Blotting.Results The pre-incubated with morroniside(1,10 and 100μmol/L)de-crease the release of Cyt C caused by H2O2-induced injury by 18%(P<0.05),28%(P<0.05),35%(P<0.01)compared to the cells exposed only to H2O2.The expression of BDNF in morroniside-treated group were higher than that of the group after injury induced by H2O2.Conclusion Morroniside shows the neurotrophic effect against the injury induced by H2O2,through inhibiting cell apoptiosis and promote the neuranagenesis.
