牛脱蛋白松质骨、胶原、BMP、RBM复合物修复骨缺损的实验研究

摘    要:目的探讨牛脱蛋白松质骨、胶原、骨形态发生蛋白(BMP)、自体红骨髓(RBM)复合物修复骨缺损的能力及其作为自体骨移植替代材料的合理性、可行性.方法在42只家兔双侧桡骨中段制备骨缺损模型,分为4组牛脱蛋白松质骨、胶原、BMP、RBM复合物植入组(A1组20侧)、单纯牛脱蛋白松质骨植入组(A2组20侧)、自体骨植入组(B1组20侧)、空白对照(B2组20侧).在2、4、8、12周分别进行大体标本观察、X线检查、病理组织学检查及生物力学测试.结果 12周时,A1组、B1组骨缺损均已完全修复,而A2组、B2组骨缺损未修复;生物力学测试术后4周A1组的最大扭矩和抗扭刚度均高于B1组,而术后8周后则无显著性差异.结论牛脱蛋白松质骨、胶原、BMP、RBM复合物具有较强的成骨能力,可修复骨缺损,并能作为自体骨移植的一种替代材料.

Abstract:Objective To investigate the ability of repairing bone defect with the compound of cattle deproteinated spongy bone,collagen,bone morphogenic protein (BMP) and red bone morrow(RBM), and the feasibility and rationality with the compounds as bone substitute material.Methods The animal models of bilateral radius bone defect were created by surgery in the adult rabbits, and they were divided into 4 group:composited bone substitue group (A 1:n=20),simple cattle deproteinated spongy bone group(A 2:n=20),autogenous bone group(A 2:n=20),autogenous bone group (B 1:n=20),control group(B 2:n=20).The effect were observed by gross,X-ray,histopathological and biomechanics examinations at 2,4,8,12 weeks after operative.Results The effect indicated that the bone defect of A 1 group and B 1 group were perfectly repaired at 12 weeks,but not with A 2 group and B 2 group.The effect of biomechanics make clear that the Mnmax and B 2 group.The effect of biomechanics make clear that the Mnmax and GIP of A 1 group is better than that of B 1 group at 4 weeks after operation,but has no significant defference between them from 8 weeks postoperative.Conclusion The compound of cattle deproteinated spongy bone,collagen,BMP and RBM possesses much bone inductive potentialities and the ability of rebuilding bone defect as well as can act as an autogenous bone substitute material.
