PBMC中HBV cccDNA与HBV宫内感染关系的研究

目的：对外周血单个核细胞(peripheral blood mononuclear cells，PBMC)中乙型肝炎病毒(HBV)共价闭合环状DNA(covalently closed circular DNA，cccDNA)进行扩增，并对其产物直接测序鉴定，以证实PBMC为HBV肝外复制的场所，从而进一步证实PBMC感染为乙型肝炎宫内感染的途径之一。方法：选择性PCR扩增PBMC中HBV cccDNA，并对1例阳性产物直接测序鉴定，测序结果与GeneBank中已知序列进行同源性比较。结果：HBsAg阳性孕妇PBMC HBV cccDNA的检出率为9．93％，新生儿PBMC HBV cccDNA的检出率为4．58％，PBMC HBV cccDNA与公布的已知序列的同源性为100％。结论：PBMC为HBV肝外复制的场所，PBMC感染很可能是乙肝宫内感染的传播机制之一。

STUDY ON THE RELATIONSHIP OF PBMC HBV cccDNA AND INTRAUTERINE INFECTION

Objective:To confirm that PBMC is the replic ating location of HBV DNA outside the liver and that the infection of PBMC is on e of the HBV i ntrauterine infecting routes by amplifying the HBV cccDNA of PBMC and confirming its sequence.Method:Selective PCR was used to amplify the HBV cc cDNA of PBMC and the HBV ccc DNA was directly sequenced with PCR products. The sequenced res u lt was compared with data in GeneBank.Results:The detection rate of PBMC HBV cccDNA is respe ctively 9.93%in pregnanting women with positive HBsAg and 4.58% in their newbo rns. The sequence of the HBV cccDNA of PBMC showed 100% identity to the reported sequence. Conclusion:PBMC is the replicating location of HBV DNA outsi de the liver and the infection of PBMC is possibly one of the HBV intrauterine i nfecting routes.
