奥沙利铂、顺铂和卡铂抗肿瘤作用的对比性研究

目的　观察奥沙利铂、顺铂和卡铂对结肠癌的抗肿瘤作用以及与 5 FU的协同作用。方法　分组选用奥沙利铂、顺铂、卡铂、5 FU、奥沙利铂 +5 FU、顺铂 +5 FU、卡铂 +5 FU作用于对数生长的结肠癌株 ,在 2 4h、4 8h后用流式细胞仪测定各组细胞的凋亡率 ;在化疗药物作用 72h后 ,用MTT法测定各组细胞的抑制率。结果　以上药物引起的凋亡率和抑制率有较大差异 ,奥沙利铂单用或联用都比其他铂类引起的凋亡率和抑制率高。结论　体外实验结果显示 ,奥沙利铂在铂类中对大肠癌有较强的抗肿瘤作用 ,同时 ,与 5 FU有较好的协同作用。

Comparative study on the antitumor effect of oxaliplatinum, cisplatinum and carboplatinum

Objective To investigate the antitumor effect of oxaliplatinum,cisplatinum and carboplatinum on human colon cancer cell line (LS114)and the synergistic effect with 5-FU.Methods The LS1114 cells in logarithmic phase were respectively treated with oxaliplatinum,cisplatinum,carboplatinum,5-FU,oxaliplatinum+5-FU,cisplatinum+5-FU or carboplatinum+5-FU.At 24th and 48th h after treatment,the apoptotic index was by using flow cytometry.At 72nd h after treatment,the inhibiting rate of the cell line was measured by MTT.Results There were difference in the apoptotic index and inhibiting rate among the above durgs.Single or combined use of oxaliplatinum could induce a higher apoptotic index and inhibiting rate than other platinum compounds.Conclusion Oxaliplatinum has strong antitumor effect against colon cancer and synergistic effect with 5-FU.
