基于苯甲酸HPLC定量分析的制草乌氨基醇型乌头原碱类水解产物的含量测定

摘    要:目的建立推算乌头碱氨基醇型水解产物含量的方法。方法通过控制苯甲酰乌头原碱水解条件,分别获得不同阶段的水解产物,确定苯甲酰乌头原碱降解率与苯甲酸转化率之间的关系,建立通过苯甲酸HPLC定量分析推导氨基醇型乌头原碱类水解产物含量的方法,并利用该方法对药典法制草乌生物碱含量进行测定。结果苯甲酰乌头原碱降解率与苯甲酸转化率约为1∶1,通过测定苯甲酸含量计算氨基醇型乌头原碱类含量的公式为:氨基醇型乌头原碱类含量≈(苯甲酸的含量×529)/122。药典法制草乌的双酯型生物碱、单酯型生物碱和氨基醇型乌头原碱类含量分别为0.0025%、0.16%和0.005%。结论通过定量分析苯甲酸含量推导乌头碱氨基醇型水解产物含量的方法可行,操作简单。

Abstract:Objective To establish a method that ct, ald calculate the content of hydrolysates of amino-alcohol-type aconitine. Methods By controlling the hydrolysis condition of benzoylaconine, hydrolysates at different stages were acquired respectively to explore the relationship between degradation rate of benzoylaconine and conversion rate of benzoic acid. By HPLC quantitative analysis of benzoic acid, the method that could calculate content of hydrolysates of amino-alcohol-type aconitine was established, which was also adopted to measure the content of alkaloid in prepared kusnezoff monkshood root. Results The ratio of degradation rate of benzoylaconine to conversion rate of benzoic acid was approximately 1 : 1. By measuring benzoic acid, the formula to calculate the content of amino-alcohol-type aconitine was : content of amino-alcohol-type aconitine~ ( content of benzoic acid ~ 52) / 122. The contents of ester-alkaloids, single ester-alkaloids and amino-alcohol-type aconitine in prepared kusnezoff monkshood root were 0. 0025%, 0. 16% and 0. 005%. Conclusion By quantitative analysis of content of benzoic acid to deduce the method that could calculate content of hydrolysates of amino-alcohol-type aconitine is feasible with convenient manipulation.
