脑卒中后大鼠边缘系统血管内皮生长因子动态变化及中药干预机制研究

摘    要:目的从脑卒中后大鼠边缘系统血管内皮生长因子(VEGF)的动态变化,探讨脑卒中后边缘系统VEGF的演变趋势及颐脑解郁方对其的早期干预作用。方法选择雄性Wistar大鼠,经Open—Field行为学评分后,随机分为正常组、假手术组、多发脑梗(MCI)模型组、血管性痴呆(VD)模型组、中药组、西药组。正常组采用常规饲养,假手术组经颈外动脉推注0.3mL生理盐水,其余各组均采用同种系微栓子体外注入法,由颈外动脉推注同等剂量的同种异体血栓栓子。VD组术后28天经水迷宫筛选后进入实验;中药组及西药组在模型制备后,分别予颐脑解郁方、尼莫地平干预给药4周;于第1、2、4周观察边缘系统VEGF的变化及药物干预效果。结果VEGF免疫组化染色结果显示,MCI模型组第1周VEGF表达明显升高,之后逐渐下降,7天为表达高峰期。VD组第1周VEGF表达低于MCI模型组,中药组第4周VEGF表达明显高于正常组及MCI模型组。结论脑卒中后大鼠存在边缘系统VEGF的改变,颐脑解郁方能加速血管的修复和再生,改善神经存活微环境而起到恢复受损功能,从而达到改善记忆和学习的功效。

Abstract:Objective To investigate the and evolutionary tendency of the vascular endothelial growth factor (VEGF) of limbic system in post stroke and the early intervention effect of Yinao Jieyu Formula from the dynamic change of VEGF of limbic system in rats with post stroke. Methods Male Wistar rats were selected and randomly divided into a normal group, a sham-operation group, multiple cerebral infarction (MCI) group, vascular dementia (VD) group, TCM group and Western medicine group according to the scores of Open-Field test. The normal group was fed routinely. The sham-operation group was injected 0. 3 mL of normal saline through external carotid artery. Homogenous microthrombus were injected into internal carotid artery in other groups. VD group received water maze screening, and the standard rats entered laboratory 28 days after operation. The TCM group and Western medicine group received Yinao Jieyu Formula and Nimodipine respectively for 4 weeks after modehng. After successful model duphcating, the evolutionary tendency of the VEGF of limbic system and the mechanism of TCM interventional effects were observed at the 1st, 2nd and 4th week. Results VEGF determination showed that MCI group had a obviously higher VEGF expression at the 1st week, which then gradually declined, and 7 days were its peak period, and. VD group had a lower expression than the MCI group at the 1 st week, but still positive ceils number was more than that in the normal group. VEGF expression of TCM group was obviously higher than that of normal group and MCI group. Conclusion Post stroke rats exist VEGF change of Ilmbic system, and the Yinao Jieyu Formula can speed up the vascular repair and regeneration, improve the microenvironment of neural survival to recover impairment function, so as to achieve the effect on improving the memory and learning.
