乙型肝炎患者细胞遗传学及淋巴细胞分裂动力学的研究

本文对乙型肝炎患者的染色体畸变、姐妹染色单体互换、核糖体核糖核酸基因(rRNA-gene)活性及淋巴细胞分裂动力进行了研究。结果表明:(1)乙型肝炎(包括无症状HBsAg镌带者、慢性迁延型肝炎和慢性活动型肝炎)的染色体畸变率及SCE率明显高于对照组,差异有高度显著性(P<0.01)。鉴于染色体畸变和SCE率的增加能反映乙型肝炎患者的肝和淋巴细胞内DNA的受损情况,故可考虑作为了解乙肝患者病情和预后,用于筛选治疗乙肝药物的检测手段。(2)乙型肝炎患者rRNA基因数目减少和转录活性降低。与对照组相比差异非常显著(P<0.01)。(3)乙型肝炎患者淋巴细胞经72小时培养进入M_3期的细胞数与对照组相比无显著差异(P>0.05),提示,细胞分裂周期并未延长。

Study on the Cytogenetics and the Lymphocytic division kinetics of patients with Hepatitis B

Chromosomal aberration, sister chromatid exchange (SCE), activity of ribosomal ribonucleic acid (rRNA) gene and lymphocytic division kinetics were studied on the patients with hepatitis B. The results indicated that (1) both of chromosomal aberration rate and SCE rate of the patients.(including HBsAg carriers who were symptomless, chronic persistant and chronic active hepatitis) are significantly higher than those of the control group (P<0.01). Increase of chromosomal aberration and the rate of SCE can reflect the damage of DNA within hepatitic cells and lymphocytes of the patients, so it can be a checkmg method to investigate the state and progrosis of the patients and to. select the drugs for'the patients with hepatitis B;(2)rRNA gene numbers of the patients were significantly lower than those of the control group(p<0.01); and (3) the numbers of third mitosis cells(72hr culture) are not significantly different between the patients and the control group(P>0.05). The fact indicated that cell cycle of patients with hepatitis B was not prolonged.
