藤茶提取物抗慢性肝纤维化作用

摘    要:目的:观察藤茶提取物(TF)抗慢性肝纤维化的作用及机制。方法:将Wistar大鼠随机分为TF高(200 mg.kg-1)、中(100 mg.kg-1)、低(50 mg.kg-1)剂量组,秋水仙碱(colchicine,Colc,0.1 mg.kg-1)组,模型组和正常组,ig给药。给药同时各组皮下注射(sc)四氯化碳(CCl4)造模,每周2次。于10周后,检测大鼠血清丙氨酸转氨酶(ALT),天冬氨酸转氨酶(AST),碱性磷酸酶(AKP),白蛋白(ALB),球蛋白(GLB),肝组织羟脯氨酸(Hyp),丙二醛(MDA),谷胱甘肽过氧化物酶(GSH-Px)及苏木精-伊红(HE)染色后的肝组织病理变化。结果:与模型组比较,TF高、中、低剂量组均能明显降低肝纤维化大鼠血清中ALT,AST,AKP,GLB的含量(P<0.01)及肝组织中MDA和Hyp的含量(P<0.05),明显提高血清ALB水平(P<0.01)及肝组织GSH-Px活性(P<0.01);肝脏组织学检查表明,TF可明显改善肝组织病理损伤程度,其作用呈剂量依赖性。结论:TF对实验性慢性肝损伤具有保护肝细胞,减少肝损伤,抗肝纤维化作用。

Abstract:Objective: To investigate the effects of Tengcha extract(TF)on hepatofibrosis induced by chronic hepatic injury. Method: Wistar rats were randomized into six groups: TF treatment (200, 100 and 50 mg ·kg-1) groups, colchicines treatment(Colc,0.1 mg ·kg-1)group, model group and normal control group. Rats were injected carbon tetrachloride (CCl4) subcutaneously two times a week to induce hepatofibrosis as well as ig administration of TF or colchicines. After 10 weeks, the levels of alanine aminotransferase(ALT),aspartate aminotransferase(AST), alkaline phosphatase(AKP), albumin(ALB), globulin (GLB) in serum and hydroxyproline(Hyp), malondialdehyde(MDA), glutathione peroxidase(GSH-Px) in liver tissue were determined, and liver histological changes were investigated by hematoxylin-eosin(HE) staining. Result: Compared with model group, TF significantly reduced the levels of ALT, AST, AKP and GLB in serum(P<0.01),and decreased Hyp, MDA level in liver tissue (P<0.05)while ALB, A/G, GSH-Px were elevated significantly(P<0.01). Pathological examination revealed that TF had therapeutical effect on experimental hepatofibrosis. Conclusion: TF has better effects on protecting hepatocyte, relieving liver injury and against hepatofibrosis.
