紫石英对排卵障碍大鼠卵巢局部卵泡刺 激素受体、黄体生成素受体表达的影响

摘    要:目的:探讨紫石英对排卵障碍大鼠模型卵泡刺激素受体(FSHR)、黄体生成素受体(LHR)的影响以及紫石英作用的物质基础。方法:选用未成年24日龄SD雌性大鼠60只随机分为模型组、克罗米芬组、紫石英组、氟化钙组、补肾活血方组、正常组6组。用Bogovich法建立大鼠排卵障碍病理模型,给药组分别ig紫石英4.8 g.kg-1,氟化钙4.0 g.kg-1,补肾活血方16.2 g.kg-1,连续14 d,克罗米芬0.009 5 g.kg-1,连续5 d。以克罗米芬及补肾活血汤为对照,用免疫组化法观察紫石英对排卵障碍大鼠卵巢局部FSHR与LHR表达的影响。结果:模型组卵巢卵泡膜细胞存在LHR高表达,与正常组比较P<0.01;卵巢颗粒细胞存在FSHR低表达,与正常组比较P<0.05。紫石英组、氟化钙组、补肾活血方组及克罗米芬组与模型组比较,卵巢组织FSHR的表达增强(P<0.05),卵巢组织LHR表达降低,但仅氟化钙组P<0.01。紫石英组与氟化钙组比较,FSHR差异无统计学意义,LHR表达强度高于氟化钙组,差异有统计学意义(P<0.01)。结论:紫石英可明显提高FSHR在卵巢颗粒细胞中的表达强度,其对FSHR的激动作用及对LHR的抑制作用可能源于紫石英中氟化钙和其他成分的共同作用。紫石英对FSHR,LHR在卵巢组织中表达水平的影响,可能是其改善大鼠排卵障碍的机制之一。

Abstract:Objective: To explore the material basis of the fluoritum's influence on the rat model with ovulation failure on FSHR and LHR. Method: Sixty 24-day-old SD female young rats were divided into six groups at random:model group,clomifene group,fluoritum group (4.8 g·kg-1),calcium fluoride group (4.0 g·kg-1),Bushen Huoxue soup group (16.2 g·kg-1) and normal control group.And using the immunohistochemical method,the level of FSHR and LHR of the rats with ovarian failure was observed. Result: FSHR in ovarian failure model rats was under-expressed and LHR was up-expressed in theca cells and granulose cells.Compared to the model group,FSHR,LHR levels in the group of fluoritum, calcium fluoride. Bushen Huoxue soup, clomifene were obviously adjusted(P<0.05 or P<0.01). Conclusion: Fluoritum can adjust the expression of FSHR,LHR in theca cells and granulose cells.Fluoritum can stimulate the follicular development and the excretion of follicle on the anovulative model rats.
