通脉复方有效组分抑制肾小球系膜细胞增殖的活性筛选及配伍研究

摘    要:目的:筛选黄芪、三七组成的通脉复方有效组分,并确定其最佳配伍剂量。方法:SD大鼠随机分为空白血清组、金水宝组1.32 g·kg-1、通脉水煎8 g·kg-1组、通脉提取物8 g·kg-1组、复方多糖0.64 g·kg-1组、复方皂苷0.16 g·kg-1组、其余组分组,共7组。ig给药7 d,7 d后采集大鼠含药血清。采用四甲基偶氮唑盐(MTT)比色法测定不同提取组分含药血清对肾小球系膜细胞(GMC)增殖的影响。在此基础上,采用2因素3水平的正交试验L9(34)对"有效组分"多糖、皂苷最佳配伍组合进行了研究。结果:正常培养及LPS刺激条件下,复方多糖与复方皂苷、通脉提取物、水煎液、金水宝含药血清均能抑制GMC增殖,与空白对照组相比,均有显著或非常显著性差异(P<0.05~0.01);而其余组分含药血清对GMC增殖无明显抑制,与空白对照组相比无显著性差异;通脉提取物含药血清对GMC增殖的抑制作用强于复方多糖、复方皂苷及金水宝含药血清,组间比较,有显著或非常显著性差异(P<0.05~P<0.01)。对主效应即GMC增殖的抑制作用的贡献程度(F值)复方多糖(13.69)与复方皂苷(13.67)几乎无差别。有效组分不同剂量配伍中,以A3B3即复方多糖960 mg、复方皂苷240 mg配伍对GMC的抑制作用最为显著。结论:复方多糖与复方皂苷为通脉复方的主要有效组分,其比例为4:1(960 mg:240 mg)为有效组分最佳剂量配伍。
