金黄色葡萄球菌对成年大鼠牙槽骨成骨细胞侵入的观察

目的了解金黄色葡萄球菌(staphylococcus aureus,SA)能否侵入体外培养的成年大鼠牙槽骨成骨细胞及其侵入能力。方法采用SA菌株ATCC 25923与体外培养的成年大鼠牙槽骨成骨细胞,按感染复数(multiplicity of infection,MOI)为100∶1的比例即100个细菌比1个细胞侵入15~60 min,通过透射电镜观察细胞内的菌落计数(colony forming unit,CFU),了解细菌侵入情况。结果透射电镜观察显示,30、45、60 min组有侵入成骨细胞现象,以60 min侵入最多,其次分别为45、30 min,SA ATCC25923对成骨细胞的15、30、45、60 min侵入菌落数分别为(13±5)、(435±31)、(803±95)和(1 800±175)CFU/孔。结论SA能侵入成骨细胞内,其侵入数量随时间延长而增多。

Observation of staphylococcus aureus invading mature rat alveolar osteoblasts

Objective To study whether staphylococcus aureus (SA) can be internalized by cultured mature rat alveolar osteoblasts(MRAOBs) and understand its abilities of invasion.Methods Staphylococcus aureus ATCC 25923 and MRAOBs were used and MRAOBs were infected at a multiplicity of infection(MOI) of 100∶1(100 bacteria∶1osteoblast) with SA for 15 to 60 min.Part of infected cells were sent for transmission electron microscope(TEM) examination and we counted intracellular bacterium colony forming unit(CFU).Results TEM showed SA were internalized at latter three time points with SA being ingested more at 60 min and followed at 45 min and 30 min.The mean ± SD CFU in one well of osteoblast invasions of SA ATCC 25923 for 15,30,45 and 60 minutes was 13±5,435±31,803±95 and 1 800±175,respectively.Conclusion The results suggested that SA ATCC 25923 could be internalized by MRAOBs.The invasion level of bacteria was based on their invasion time.
