半夏栽培品遗传差异的AFLP分析

摘    要:目的构建不同地区来源半夏栽培品的DNA指纹图谱,探讨利用AFLP分子标记在半夏遗传多样性、亲缘关系及种质鉴别上的可行性。方法运用扩增片段长度多态性技术(AFLP),对我国17个地区的51份半夏栽培品和4份掌叶半夏样品(外群)进行基因组DNA多态性分析。结果从64对引物组合中筛选出8对多态性丰富、鉴别效率高的AFLP引物组合,构建了51份半夏栽培品的AFLP指纹图谱。聚类分析结果表明:所有半夏栽培品完全被区分开,来源地相同的种质表现出相对密切的亲缘关系,浙江等华东地区的栽培半夏同其他地区半夏有着明显遗传差异,聚类分析结果获得了Bootstrap校验的支持。结论AFLP分子标记可用于半夏遗传多样性、亲缘关系及种质鉴别分析,浙江、江苏等华东地区栽培半夏的遗传特性相对独立。

Abstract:Objective The DNA fingerprints of cultivated Pinellia ternata collected from different geographic regions were generated by using AFLP markers to find the feasibility in analyzing their genetic diversity, relationship, and germplasm identification. Methods The DNA polymorphism of 51 cultivated germplasm of P. ternata collected from 17 different habitats and four cultivars of P. pedatisecta (outgroup) were detected by AFLP molecular markers. Results The DNA fingerprints of 51 individuals of P. ternata were obviously distinguished by eight pairs of high polymorphic and efficient primer combinations screened from 64 primer combinations. The phylogenetic clustering results revealed that all the tested cultivars were fully differentiated, and individuals from the same regions were mainly clustered together. Moreover, cultivars from East-China, including Zhejiang and Jiangsu Provinces, displayed clear genetic distinction from other regions. The clustering results were strongly supported by Bootstrap test. Conclusion AFLP Markers can be potentially used in analyzing of genetic diversity, relationship, and germplasm identification of this medicinal plant, and the germplasm from regions of East-China, including Zhejiang and Jiangsu Provinces, displays the relative separate genetic characters from other regions.
