结缔组织生长因子、血管内皮生长因子、缺氧诱导因子-1α在胰腺癌中的表达及临床意义

摘    要:目的:检测胰腺癌中CTGF、VEGF、HIF-1α的表达情况,探讨三者在胰腺癌缺氧微环境中对胰腺癌发生、发展的作用。方法:采用免疫组化Envision二步法对46例胰腺癌组织和10例癌旁胰腺组织中CTGF、VEGF、HIF-1α的表达进行检测。结果:CTGF、VEGF和HIF-1α在胰腺癌组织中均高表达,阳性率分别为45.7%、71.7%、76.1%,显著高于三者在癌旁胰腺组织的表达(0)(P〈0.01)。CTGF在胰腺癌组织中的表达水平与性别、肿瘤部位无关,与年龄、临床分期、病理分级、淋巴结转移情况有关(P〈0.05)。VEGF和HIF-1α在胰腺癌组织中的表达水平只与临床分期有关(P〈0.05),与其他肿瘤生物学行为无关。CTGF与VEGF、HIF-1α,VEGF与HIF-1α之间的Spearman相关系数分别为r=0.575、r=0.483、r=0.082,即三者两两间均存在正相关关系(均P〈0.01)。结论:胰腺癌组织中CTGF、VEGF、HIF-1α的表达与在癌旁胰腺组织中的表达有显著差异,三者对胰腺癌在肿瘤缺氧微环境下的发生、发展有重要作用。

Abstract:Objective: To investigate the expressions of CTGF,VEGF,HIF-1α and the relationships between them and various clinical pathological characteristics in human pancreatic carcinoma.Methods: To investigate the expressions of CTGF,VEGF and HIF-1α in tumor issues from 46 radically resected specimens of pancreatic carcinoma and 10 normal pancreatic tissues by Envision two-step immunohistochemical staining.Results: CTGF,VEGF and HIF-1α in pancreatic cancer tissues are high expressed,significantly higher than that in normal pancreatic tissues.The Positive rate of CTGF was closely related to the age,clinical stage,pathologic stage grading and lymph node metastasis(P0.05).The Positive rates of VEGF and HIF-1α was closely related to the clinical stage(P0.05),and other tumor biological behaviour was irrelevant.The spearman correlation coefficients of CTGF and VEGF,HIF-1α,VEGF and HIF-1α were r=0.575,r=0.483l,r=0.082 respectively.Positive correlations were mutually found between each two of the three(P0.01).Conclusion: Up-regulated expressions of CTGF,VEGF and HIF-1α exist in human pancreatic carcinoma.CTGF,VEGF and HIF-1α play an important role in the occurrence and development process of the pancreatic cancer in the hypoxia microenvironment.
