胶体对大鼠肝脏低温保存及移植后微循环保护作用的研究

摘    要:灌洗保存液中肢体渗透压的维持被认为是器官保存的必要条件之一。本研究应用大鼠原位肝移植模型采用生化检测、共焦激光扫描显微镜技术研究发现,保存液中加入羟乙基淀粉(HghroxyethylStarchHES)或低分子石旋糖酐Dex40能够有效地防止肝脏低温保存的肝血窦内皮细胞损伤,同时能改善移植后再灌注肝脏的微循环障碍,进而保护移植肝功能、DeX40的作用优于HES。结论:保存液中加入胶体成分,对防止肝脏的缺血再灌注损伤是有益的,可能与其能有效地维持胶体渗透压有关。Dex40可成功地替代HES作为维持胶体渗透压的有效成分。

Abstract:The maintenence of colloid osmorlarity pressure in per fusion and preservation fluid is thoughtas one key point in organ preservation. Orthotopic liver transplantation in rats was processed and both biochemical test and confocal laser scan microscopy were used in our study. The result shows: Adding HES orDex40 into preservation fluid can prevent sinusoidal lining cell damage following cold preservation effectively,and improve the hepatic microcirculation disturbancc following transplantation and reper fusion,thus protecting the function of transplanted liver. Conclusion:Adding Colloid into preservation fluid is beneficial to prevent ischemic reperfusion damage of the liver,which may be related to it's effect of maintaining the colloid osmorlarity pressure. ixxtran40 may be an ideal substitue of HES in the preservation solution,since it's more economic and effective.
