| 摘 要： | ①目的 对发现于招远市的异常血红蛋白(Hb)进行分子结构的研究。②方法 取异常Hb血样,以生理盐水洗去血浆蛋白质,再以蒸馏水处理压积红细胞以溶血,溶血液以-20℃的酸性丙酮处理提取珠蛋白,用尿素将珠蛋白解链,再经柱层析分离及纯化异常链。③结果 经羧甲基纤维素及交联葡聚糖层析分离及纯化得到异常α或β链,用十二烷基硫酸钠-聚丙烯酰胺凝胶电泳(SDS—PAGE)检查纯度,这些异常α或β链均为一条电泳带。④结论 纯化的异常α或β链均电泳纯净。

| Abstract: | Objective To separate and purify the abnormal hemoglobins(Hb) found in Zhaoyuan. Method The abnormal Hb blood samples were washed with normal saline and distilled water. The resulted hemolysis solution was treated with acidic acetone at -20C to obtain the globin which was later dissociated by urea, separated and purified with.column chromatography. Result After separation and purification of the abnormal o or p chains with CM-cellulose and sephadex column chromatography, we examined their purities with SDS-PAGE. The abnormal o and P chains showed each one single band upon electrophoresis. Conclusion The purified abnormal a or fl chains are pure in electrophoresis. |
