胡桃叶黄酮对D-半乳糖衰老模型小鼠的影响

摘    要:目的 观察胡桃叶总黄酮对d-半乳糖衰老模型小鼠的SOD、ACHE、MDA的影响,探讨其抗衰老作用及机制.方法 小鼠随机均分为6组,除正常对照组外,各组每天皮下注射D-半乳糖(0.05g/kg),连续注射6周.从注射的第7天开始,3个实验组同时灌胃高、中、低剂量的胡桃叶总黄酮,阳性对照组灌以维生素E(0.05g/kg)模型对照组与正常对照组每天灌胃等量的蒸馏水,连续给药5周后测定血清SOD、ACHE和脑组织MDA的含量变化.结果 实验组的血清SOD显著高于模型对照组(P<0.05),ACHE活力极显著高于模型对照组(P<0.01),实验组的脑MDA含量显著低于模型对照组(P<0.05);与正常对照组比,各项指标相近(P>0.05).结论 胡桃叶总黄酮能提高抗氧化能力,降低自由基含量,增强胆碱能神经系统功能,从而防止衰老.

Abstract:Objective To observe the effects oftotal flavone of Juglans regia folium on superoxide dismutase(SOD),ACHE,malondiadehyde(MDA)in mice to study therapeutic effects and mechanism of Total flavone of Juglans regia folium on doddery models created by D-galactose. Method Mice were randomly divided into 6 groups. All groups,except the control,were administrated(ip) with D-galactose(0.05g/kg) for successive 6 weeks. The 3 experimental groups were administrated(ig) with high,middle and low dosages oftotal flavone of Juglans regia folium respectively after 7th day of the injection of D-galactose whereas vitamin-E at 0.05g/kg body weight served as a positive control. The model and control groups were daily adminisered with D.D.W. for continuous 5 weeks. After treatments,the SOD,ACHE activity in serum and MDA in brain of rats were determined. Result The serum SOD,ACHE activities in total flavone of Juglans regia folium -treated mice were higher than that in control group. However,the brain MDA content in total flavone of Juglans regia folium-treated mice was lower than that in control group.Conclusion This data suggested thattotal flavone of Juglans regia folium had a significant anti-oxidative effect by reducing the free radicals as well as an anti-aging effect through the enhancement of cholinergic nerve system.
