六味地黄汤不同配伍对马钱素药物动力学的影响

目的研究六味地黄汤不同配伍对马钱素药物动力学的影响。方法小鼠一次灌胃六味地黄汤、三补汤(山茱萸、山药、地黄)、一补一泄汤(山茱萸、丹皮)、山茱萸汤后,用建立的 HPLC 法测定吸收入血的马钱素血药浓度-时间曲线,用梯形法计算血药浓度-时间曲线下面积(AUC),比较马钱素的吸收速度及程度。结果以 C_(max)/AUC、T_(max)和 t_(1/2ka)为吸收速度指标,以一补一泻汤最快,其次为三补汤及全方汤,单味药山茱萸汤最慢;以 AUC 及 C_(max)为吸收程度指标,单味药山茱萸汤吸收程度最高,一补一泻汤最低。结论六味地黄汤不同配伍会影响马钱素药物动力学。

Effect of Liuwei Dihuang Decoction with Different Combinations on Pharmacokinetics of Loganin

Objective To observe the effect of Liuwei Dihuang decoction(LDD)with different combinations on pharmacokinetics of loganin.Methods Mice were administered once with LDD and its different combinations,that were Sanbu decoction(A,contained dogwood fruit,Yam rhizome and rehmannia root),Yibu Yixie decoction(B,contained dogwood fruit and moutan bark)and Shanzhuyu decoction(C,contained dogwood fruit).Then the concentration-time curve of loganin absorbed into blood was performed by established HPLC method,and the area under curve(AUC)was calculated to compare the velocity and degree of loganin absorption.Results In regard to the velocity of absorption, taking C_max/AUC,T_(max) and t_1/(2ka)_as the indexes,decoction B was the quickest one,decoction A and LDD the second,and decoction C the slowest.In regard to the degree of absorption,taking AUC and C_(max) as the indexes,decoction C was the highest and decoction B the lowest.Conclusion Different combinations of LDD could affect the pharmacokinetics of loganin in different manner.
