复方氨基酸(15)双肽(2)注射液中两组分的含量测定

摘    要:目的建立复方氨基酸(15)双肽(2)注射液中两种二肽的含量测定方法。方法以2,4-二硝基氟苯为衍生剂进行柱前衍生,以C18柱为固定相、乙腈-磷酸盐缓冲液为流动相进行梯度洗脱,于360nirl处检测。结果所有组分于45min内洗脱完毕,N-甘氨酰-L-谷氨酰胺回收率为99.8%,RSD为2.63%;N-甘氨酰-L-酪氨酸回收率为100.9%,RSD为3.67%。结论该方法操作简便,结果准确,可用于复方氨基酸(15)双肽(2)注射液中N-甘氨酰-L-谷氨酰胺和N-甘氨酰-L-酪氨酸的含量测定。

Abstract:Objective To establish a method for the determination of N- glycyl- L- glutamine and N- glycyl- L- tyrosine in Compound Amino Acids (15) and Dipeptides (2) Injection. Methods The amino acids and dipeptides reacted with FDNB respectively and quantitatively and produced the corresponding derivatives which were analyzed by HPLC. Chromatographic conditions were as follows: C18 column,the mobile phase of acetonitrile-phosphate buffer solution,360 nm as the detective wavelength. Results All components were completely eluted in 45 min. The recovery rate of N-glycyl-L-glutamine and N-glycyl-L-tyrosine was 99.8% and 100.9%, RSD 2.63% and 3.67%, respectively. Conclusion The method is simple, accurate and suitable for the determination of N- glycyl- L-glutamine and N- glycyl-L-tyrosine in Compound Amino Acids (15) and Dipeptides (2) Injection.
