香丹注射液抗急性脑缺血细胞间粘附作用的实验研究

摘    要:目的 探讨香丹注射液在急性脑缺血后的抗细胞间粘附作用。方法 成年♂Wistar大鼠 2 4只 ,随机分为正常对照组、伪手术组、脑缺血 -再灌注损伤组 (脑缺血组 )、脑缺血 -再灌注损伤加香丹注射液组 (脑缺血加香丹注射液组 ) 4组 ,每组 6只大鼠。分离培养大鼠脑毛细血管内皮细胞 (CCEC)和多形核白细胞 (PMN) ,利用微管吸吮技术 ,观察各组CCEC和PMN间粘附力特性的变化。结果 脑缺血 -再灌注后各时间点CCEC和PMN的粘附力和粘附应力均明显高于正常对照组和伪手术组 (P <0 .0 1 ) ;加香丹注射液后 ,CCEC和PMN的粘附力和粘附应力均明显下降 (P <0 .0 5或P <0 .0 1 )。结论 脑缺血 -再灌注损伤后 ,香丹注射液可抑制CCEC和PMN的粘附 ,减轻脑损伤。

Abstract:OBJECTIVE To explore the effects of Xiangdan injection on intercellular adhesion in acute cerebral ischemia in rats.METHODS 24 male Wistar rats were randomly divided into 4 groups: control group (CG),shamed operative group (SOG), cerebral ischemia-reperfusion injuried group (CIG) and cerebral ischemia-reperfusion injuried and cured by Xiangdan injection group (CTG),and each group had 6 samples.Cerebral capillary vessel endothelial cell (CCEC) and polymorphism nuclear leukocytes (PMN) in rats were separated and cultivated, and the changed of characters of CCEC and PMN were observed in every group by means of microburet suck technique.RESULTS Compared CG with SOG, the adhesion force and adhesion stress of CCEC and PMN in the sample of CIG and CTG were higher, and there were significant difference( P < 0.01 ) in different time point. After Xiangdan injection perfusion , the adhesion force and adhesion stress of CCEC and PMN in the samples of SOG and CTG were reduced, and there were significant difference( P <0.05 or P <0.01).CONCLUSION Xiangdan injection can inhibit adhesion function of CCEC and PMN and alleviate cerebral injury after ischemia-reperfusion
