实时荧光核酸恒温扩增技术在淋球菌检测中的应用分析
摘    要:目的采用统计学方法评价实时荧光核酸恒温扩增技术(SAT)在临床淋球菌检测中的应用。方法以淋球菌培养法为标准对照,采用实时荧光核酸恒温扩增技术同时对150例疑似患者的生殖道拭子标本和尿液标本进行检测,对结果进行统计学分析比较,并用PCR法进行验证。结果使用SAT检测的生殖道拭子和尿液阳性检出均为110例,培养法阳性检出为108例,其中培养法检出阴性的2例通过PCR验证为阳性。两种取样方式的SAT检测法和培养法三者阳性率差异均无统计学意义(P0.05);以培养法为金标准,SAT检测拭子的敏感度为100.0%,特异度为95.2%;SAT检测尿液标本的敏感度为100.0%,特异度为95.2%。结论采用实时荧光核酸恒温扩增技术对生殖道拭子和尿道标本的检测效果与培养法相比,阳性率高,敏感度较高,且尿道标本收集方法比较简便,患者依从性好,可代替拭子道取样方法,此方法可在临床推广应用。

Application analysis of real-time fluorescent nucleic acid constant temperature amplification technology in detection of neisseria gonorrhoeae
Abstract:Objective To evaluate the application of real-time fluorescent nucleic acid constant temperature amplification(SAT)in the detection of clinical gonococcal bacteria by statistical methods.Methods The neisseria gonococcal culture method was used as the standard control.The genital swab specimens and urine specimens of 150 suspected patients were detected by real-time fluorescent nucleic acid constant temperature amplification technique.The results were statistically analyzed and compared,and were verified by PCR.Results The positive detection of genital swabs and urinary tracts detected by SAT was 110 cases,and the positive detection by culture method was 108 cases.Among them,2 cases with negative detection by culture method were positive by PCR.There was no significant difference in the positive rate between the two methods of SAT and culture method(P>0.05).Using the culture method as the gold standard,the sensitivity of SAT was 100.0%and the specificity was 95.2%.The sensitivity of the SAT for urine samples was 100.0%and the specificity was 95.2%.Conclusion The detection effect of the real-time fluorescent nucleic acid constant temperature amplification technology on the genital swab and urethral specimens is higher than that of the culture method.The positive rate is high,the sensitivity is high,and the urinary tract specimen collection method is relatively simple.The patient compliance is good and can be replaced with swab sampling method.The real-time fluorescent nucleic acid constant temperature amplification technology can be applied in clinical practice.
