| 摘 要： | 目的 研究三氧化二砷(As2O3)对人大肠癌细胞株LoVo中的生长及三氧化二砷对Bcl-2、Bax表达的影响.方法 采用不同浓度的As2O3处理LoVo细胞,MTT法测定细胞的生长抑制效应;免疫细胞化学法检测人结肠癌细胞LoVo中Bcl-2、Bax的表达.结果 MTT结果示As2O3对大肠癌LoVo细胞的生长有抑制作用,且有时间-剂量依赖性(P<0.05或P<0.01);免疫细胞化学法结果示As2O3可使LoVo细胞中Bcl-2表达下降,Bax表达升高(P<0.01).结论 三氧化二砷对大肠癌细胞生长有抑制作用,且呈时间、剂量依赖性;其机制可能与改变Bcl-2和Bax的表达水平有关.

| Abstract: | Objective To investigate the effects of arsenic trioxide on the cell growth and the expression of Bcl-2 and Bax in human colorectal cancer cells.Methods In vitro,human colorectal cancer cells was treated by As2O3 in various concentrations,and then MTT method was used to assay the effect of cell growth inhibition;the immunocytochemistry(ICC) analyzed the expression of Bcl-2 and Bax.Results As2O3 inhibited the growth of the human colorectal carcinoma cells,and the inhibition effect was time-dose dependent(P<0.05 or P<0.01). Bcl-2 expression was downregulated,and Bax expression was upregulated(P<0.01).Conclusion Arsenic trioxide had inhibition effect on the growth of human colorectal cancer cells in a time-dose dependent manner, which may change the Bcl-2 and Bax expression and the level. |
