辛伐他汀预防激素性骨质疏松实验研究的初步报告

摘    要:目的 :观察辛伐他汀对糖皮质激素性骨质疏松的预防作用。方法 :取健康 6月龄SD雄性大鼠 3 6只 ,体重 0 .45± 0 .0 2kg ,随机分为对照组 (A组 )、模型组 (B组 )和实验组 (C组 ) ,每组 12只。对照组灌胃法给予生理盐水 2ml/(kg·d) ,每周 2次肌肉注射生理盐水 ,每次 0 .5ml/kg ;模型组灌胃法给予生理盐水 2ml/(kg·d) ,每周 2次肌肉注射地塞米松 ,每次 2 .5mg/kg ;实验组灌胃法给予辛伐他汀 10mg/(kg·d) ,每周 2次肌肉注射地塞米松 ,每次 2 .5mg/kg。 8周后对各组大鼠的第 3腰椎行骨密度测量和组织形态显微结构观察以及血清钙、磷和碱性磷酸酶 (ALP)含量测定。结果 :实验组第 3腰椎骨密度值明显高于模型组 (P <0 .0 1)。对照组的骨密度值与实验组比较差异无显著性意义 (P >0 .0 5 )。实验组第 3腰椎骨小梁形态结构与对照组相似 ,模型组呈骨质疏松特征。模型组ALP较对照组及实验组均明显升高 (P <0 .0 1)。结论 :辛伐他汀可有效预防糖皮质激素性骨质疏松。

Abstract:Objective: To study the effectiveness of simvastatin on preventing glucocorticoid-induced osteoporosis.Methods: Thirty-six male SD rats, weighted 0.45±0.02 kg each, were randomly divided into 3 groups (12 rats for each group):Group A, given 0.9% natrium chloride 2 ml/(kg·d) through gastric tube and 0.5 ml/kg twice a week through intramuscular injection; Group B, given 0.9% natrium chloride 2 ml/(kg·d) through gastric tube and dexamethasone 2.5 mg/kg twice a week through intramuscular injection; Group C, given simvastatin 10 mg/(kg·d) through gastric tube and dexamethasone 2.5 mg/kg twice a week through intramuscular injection. After the experiment had lasted for 8 weeks, the third lumbar vertebral bodies of all the rats were managed to do the measurement of bone mineral density and the structure of bone trabeculae,and the levels of Ca 2+ ,PO 3- 4 and ALP in serums were measured.Results: The values of bone mineral density in Group C were significantly higher than that of Group B,(P<0.01). No significant defference was observed in bone mineral density between Group A and Group C (p>0.05). The structure of bone trabeculae of Group C was similar to that of Group A, and Group B presented the feature of osteoporosis. ALP levels of Group B were significantly increased compared to the corresponding value of Group A and Group C(P<0.01).Conclusion: Simvastatin can prevent the osteoporosis induced by glucocorticoid.
