体外培养骨髓源性神经干细胞的细胞遗传学分析

摘    要:目的评价体外培养的成人骨髓源性神经干细胞的遗传稳定性。方法制备骨髓源性神经干细胞的染色体标本,以常规染色和G显带进行染色体的核型分析。结果体外培养的成人骨髓源性神经干细胞的染色体数目为正常人类的46条染色体,未见非整倍体染色体像;G显带图像核型分析显示其为二倍体正常核型,染色体中未发现有缺失、倒位、易位、双着丝粒和环状染色体等结构异常。结论体外培养分化形成的骨髓源性神经干细胞保持了遗传学上的稳定性.为其进一步的临床应用提供了实验依据。

Abstract:Objective To evaluate the genetic stability of adult neural stem cells from bone marrow cultured in vitro. Methods Chromosome preparations of neural stem cells from bone marrow were made and analyzed on karyotypes using conventional staining and G-banding techniques. Results Chromosome number of neural stem cells from bone marrow was 46 as a normal human has and there was no aneuploid. The karyotypic analysis showed normal diploid, and no such chromosome structural abnormalities as deletion, inversion, translocation, dicentric and ring chromosome. Conclusion The bone marrow-derived neural stem cells cultured in vitro preserve cytogenetic stability, which provides experimental proof to bring this kind of cells into clinic application.
