通腑法通腑利肺作用机制的实验研究

摘    要:目的 从整体、细胞及分子3个层面探讨通腑法通腑利肺的作用机制。方法 将40只SD大鼠随机等分为正常对照组、模型组、解扎组及治疗组,每组各10只,采用体外直肠不全结扎法造模。然后用放射免疫分析法检测各组血清IL-8含量,RT-PCR法检测肺组织TNFα mRNA水平,检测各组支气管肺泡灌洗液(bronchoalveolar lavage fluid,BALF)中肺泡巨噬细胞(pulmonary alveolar macrophage,PAM)死亡百分率,并将通腑治疗组、解扎组与模型组及正常对照组对照。结果 模型组血清IL-8含量明显高于正常对照组(P〈0.01),肺组织TNF-α mRNA表达明显增高(P〈0.01);通腑法治疗后IL-8含量和TNF-α mRNA表达均低于模型组(P〈0.01)。且肺损伤程度减轻。结论 IL-8、TNF-α在肠源性肺损伤过程中起重要作用。通腑法对损伤肺组织起保护作用。

Abstract:[Objective] To explore the mechanism of protective effect on the lung by the method of Tongfu through the three levels of general, cell and molecular. [Methods] Forty SD rats were randomly divided into four groups: normal control group, model group, ligation group and treatment group (n=10 each). The models were made by the method of incomplete ligating the rectum outside the body. The content of IL-8 in blood serum of each group was measured by the method of radioactive immunity. The expression of TNF-αmRNA in lung tissues was detected by RT-PCR method, and the percentage of the dead pulmonary alveolar macrophage (PAM) in the bronchoalveolar lavage fluid (BALF) was detected. In each group, the results were compared among the four groups. [Results] The content of IL-8 in blood serum and the expression of TNF-α mRNA in lung tissues in the model rats were significantly higher than those in the normal control rats (P <0.01). After the treatment of Tongfu, all of the indexes decreased and the results were lower than those in the model rats, and damage of the lung in the treatment group was reduced. [Conclusion] IL-8 and TNF-α play an important role during the process of intestinal endotoxemia caused lung damage, the method of Tongfu will protect the lung from damage.
