诱导生物合成金属硫蛋白对顺铂引起血液毒性的影响

摘    要:顺铂对小鼠血液系统具有毒性作用,而组织中金属硫蛋白(metallothionein,MT)的产生能减轻其毒性反应。本实验观察了甘草锌(Gly-Zn),富硒麦芽(Se-malt)和亚硒酸钠(Na_2SeO_3)等含微量元素的药物对诱导组织产生MT的作用及对顺铂引起血液系统毒性的影响。结果表明:富硒麦芽的MT诱导作用最强,其次为亚硒酸钠和甘草锌。甘草锌、富硒麦芽和亚硒酸钠对顺铂引起的小鼠血液系统毒性具有明显的保护作用,由此提示了在应用含锌、硒等微量元素药物降低顺铂引起重金属中毒时,诱导生物合成MT增加,可能是其中重要的机理之一。

Abstract:Cisplatin can induce hematological toxicity, and induction of metallothionein(MT) synthe-sis in tissue can reduce the toxicity. The protective effect of trace elemental compounds [Zinc glycyrrhizate (Gly-Zn), Se-malt and Na2SeO3] on inducing MT synthesis and hematological toxicity-induced by cisplatin was investigated in mice. The results showed that Se-malt had the highest activity in inducing MT synthesis, and this activity was also possessed by Gly-Zn or Na2SeO3. The compounds possessed a significant protective effect on hematological toxicity in-duced by cisplatin. It suggested that increased MT synthesis induced by the trace element compounds (Se and Zn etc) may be one of the important mechanisms to reduce the blood toxicity of cisplatin.
