人参二醇组皂甙对肾缺血再灌流性血清致伤的肾小管细胞膜稳定性的影响及机制研究

摘    要:目的:用培养的人肾小管细胞,观察人参二醇组皂甙(PDS)对肾缺血再灌流性血清致伤的小管细胞膜稳定性的影响,并探讨其机制。方法:提取兔肾缺血再灌流后血清(SIR)及对照血清(SC),用不同的培养基(普通培养基、对照血清、缺血血清以及缺血血清加PDS)分别与肾小管细胞共培养96小时,生化法测定培养液或细胞中MDA含量以及SOD、LDH、Na -K -ATP酶和Ca2 -ATP酶活力。结果:SIR可使培养液中提示细胞膜脂质过氧化程度的MDA含量和表示膜完整性的细胞外LDH活力均显著升高,同时胞内的SOD、Na -K -ATP酶和Ca2 -ATP酶活力明显降低,而PDS则可抑制SIR的这些效应的发挥。结论:PDS具有增强急性肾缺血再灌流性兔血清致伤的培养人肾小管细胞膜稳定性的作用。其机制可能为增强SOD活力、减轻氧自由基对细胞膜的攻击,增强细胞膜上ATP酶的活性以维持细胞内外水、电解质平衡等作用有关。

Abstract:Objective:To observe the effects of Panaxadiol Saponins(PDS) on the membrane stability of cultured renal tubular cells injured by the serum of renal ischemic reperfusion(IR),and discuss their mechanisms.Methods:Firstly,acquire the serum of rabbits with renal IR(SIR) and the control serum of rabbits(SC),to take the different culture mediums(common culture mediums?SIR?SC and SIR PDS)in diverse groups,and culture renal tubular cells for 96 hours,then,dete of the content of alondialdehyde (MDA) and the activity of Superoxide Dismute(SOD),lactate dehydrogenase(LDH),ATPase in the culture medium or cells by biochemistry method.Results:The content of MDA in the culture medium,showing the degree of lipid Superoxide reaction,and LDH in the culture medium,indicating the integrality of plasmalemma,were significantly increased,but the activity of SOD,Na~ -K~ -ATPase and Ca~(2 )-ATPase in cells were remarkably decreased after treated by SIR.However,these effects of SIR could be inhibited by PDS.Conclusions:The membrane stability of cultured renal tubular cells injured by the serum of renal IR could be promoted by Panaxadiol Saponins.The possible mechanisms could be concluded as follows increasing the activity of SOD,and relieveing the injury of membrane resulting from the attacking of Oxygen free radical,increasing the activity of ATPase and maintaining the balance of the content of water and electrolyte inside and outside the cells.
