不同剂量乌司他丁对急性百草枯中毒大鼠过氧化损伤的保护作用

摘    要:目的:观察不同剂量乌司他丁(UTI)对急性百草枯(PQ)中毒大鼠过氧化损伤的保护作用。方法将50只SD大鼠随机分为5组,正常对照组(A)、PQ中毒组(B)、小剂量UTI治疗组(C)、中剂量UTI治疗组(D)、大剂量UTI治疗组(E),每组10只。 B、C、D、E组大鼠采用PQ一次性灌胃染毒法(80mg/kg)复制PQ中毒肺损伤模型,A组采用等量生理盐水灌胃。灌胃30min后,C组腹腔注射UTI100KU/Kg,D组腹腔注射UTI300KU/Kg,E组腹腔注射UTI600KU/Kg,A、B组注射等量生理盐水。24h后检测血清丙二醛(MDA)、超氧化物歧化酶(SOD)浓度,观察各组大鼠的一般情况,蛋白含量变化。光镜下观察肺组织病理学变化,进行肺损伤评分。结果 C、D、E组和B组比较,大鼠血清MDA、SOD浓度,肺组织损伤病理评分存在显著差异(P〈0.05),C、D、E组之间比较也有显著差异(P〈0.05)。结论大剂量UTI对急性PQ中毒大鼠过氧化损伤具有较强保护作用。

Protective effect of different dose of ulinastatin on acute peroxidation injury induced by paraquat-intoxicated in rats

Abstract:Objective To observe the protective effect of different dose of ulinastatin (UTI) on acute peroxidation injury in-duced by paraquat(PQ)-intoxicated in rats. Methods 50 SD rats were randomly divided into 5 groups,normal control group(A),PQ poisoning group(B),low dose UTI treatment group(C),middle dose UTI treatment group(D),high dose UTI treatment group(E),10 rats in each group. B,C,D,E group of rats were made into the model of lung injury by intragastric PQ (80mg/kg)administration, and same quantity of physiological saline was administered intragastrically in group A. 30 minutes after PQ administration ,group C were intraperitoneally injected with UTI(100KU/kg),group D were intraperitoneally injected with UTI(300KU/kg),group E were in-traperitoneally injected with UTI (600KU/kg),and then the A,B group were injected same quantity of physiological saline. Observ-ing the general condition of rats after 24 hours,measuring the levels of serum MDA,SOD concentration,observing the pathological changes of lung tissues,lung injury score under the light microscope. Results Compared with B group,there were significant dif-ferences in C,D,E group with the serum MDA,SOD concentration and lung tissue pathological score (P〈0.05),and then there were significant differences between C,D,E group(P〈0.05). Conclusion High dose UTI has strong protective effect on acute per-oxidation injury induced by paraquat-intoxicated in rats.
