微小RNA-497在恶性胶质瘤组织的表达及临床意义

摘    要:目的探讨恶性胶质瘤组织中微小RNA(miR)-497表达特性及临床预后的关系。方法2015年1月至2019年7月收集南方医科大学附属珠江医院25例胶质瘤组织,16例非肿瘤病变正常脑组织组织,分为实验组及对照组。荧光实时定量聚合酶链反应(PCR)测定胶质瘤组织和正常脑组织中miR-497、Wnt3a表达,线性相关分析miR-497与Wnt3a之间的关系。根据miR-497表达量高低分为高表达组、低表达组,生存分析(Kaplan-Meier分析)无病生存曲线和总生存曲线,分析miR-497表达与胶质瘤预后之间的关系。两独立样本t检验比较胶质瘤组织和非肿瘤正常脑组织中miR-497平均表达水平;采用单因素方差分析比较实时荧光定量PCR检测结果中各个细胞株ΔCt值的差异(One-way ANOVA)。结果实时荧光定量PCR检测胶质瘤组织中miR-497的表达(1.29±0.61)低于正常脑组织(5.76±1.45,t=3.215,P<0.05),差异有统计学意义。胶质瘤组织中miR-497和Wnt3a基因RNA之间表达明显相关,miR-497和Wnt3a基因表达呈负相关(P<0.05)。高表达miR-497组与低表达miR-497组胶质瘤患者的Kaplan-Meier无病生存曲线和总生存曲线,miR-497表达高的胶质瘤患者无病生存率和总生存率显著高于miR-497低表达者,差异有统计学意义(t=5.147,P<0.05)。结论恶性胶质瘤中miR-497表达下调,Wnt3a表达上调。miR-497表达高的胶质瘤患者无病生存率和总生存率显著高于miR-497低表达者。miR-497表达水平可能与恶性胶质瘤预后明显相关。

Abstract:Objective To study the expression of microRNA(miRNA,miR)-497 in malignant glioma and its relationship with clinical prognosis.Methods From January 2015 to July 2019,25 cases of glioma tissues and 16 cases of normal brain tissues without tumor lesions were collected in Zhujiang Hospital Affiliated to Southern Medical University.They were divided into experimental group and control group.The expression of miR-497 and Wnt3a in gliomas and normal brain tissues was detected by real-time fluorescence quantitative polymerase chain reaction(PCR).The relationship between miR-497 and Wnt3a was analyzed by linear correlation.According to the expression level of miR-497,the patients were divided into high expression group and low expression group.Kaplan Meier analysis was used to analyze the disease-free survival curve and overall survival curve,and the relationship between the expression of miR-497 and the prognosis of glioma.The average expression level of miR-497 in glioma tissues and non-tumor normal brain tissues was compared by two independent samples t-test,and the difference ofΔCt value of each cell line detected by real-time fluorescent quantitative PCR was compared by one-way ANOVA.Results The expression levels of miR-497 in glioma and normal brain tissues were(1.29±0.61)and(5.76±1.45),respectively.The expression of miR-497 in glioma was lower than that in normal brain tissues(t=3.215,P<0.05).There was an obvious correlation between the expression of miR-497 and Wnt3a mRNA in gliomas,but there was a negative correlation between the expression of miR-497 and Wnt3a gene(P<0.05).For Kaplan Meier disease-free survival curve and overall survival curve of glioma patients with high expression of miR-497 and low expression of miR-497,the disease-free survival rate and overall survival rate in glioma patients with high expression of miR-497 were significantly higher than those with low expression of miR-497(t=5.147,P<0.05).Conclusion The expression of miR-497 was down-regulated and Wnt3a was up-regulated in gliomas.The disease-free survival rate and overall survival rate of glioma patients with high expression of miR-497 were significantly higher than those with low expression of miR-497.The expression of miR-497 may be obviously related to the prognosis of malignant glioma.
