真核翻译起始因子5A2和热休克蛋白90在胆囊癌组织的表达及其临床意义

摘    要:目的探讨真核翻译起始因子5A2(EIF5A2)和热休克蛋白90(HSP90)在胆囊癌组织中的表达和临床意义。方法收集2014年3月至2020年10月山西省人民医院70例胆囊癌患者癌组织标本,同时选取因结石切除的35例胆囊组织标本为对照组,采用免疫组织化学法检测上述组织中EIF5A2蛋白和HSP90蛋白水平,进行χ2检验统计学分析两者的表达与胆囊癌临床病理学参数之间的关系。结果胆囊癌组织中EIF5A2阳性表达率明显高于正常胆囊组织EIF5A2阳性表达率72.86%(51/70)比8.57%(3/35),χ2=38.603,P<0.01],差异有统计学意义,EIF5A2表达水平与胆囊癌患者淋巴结转移、TNM分期明显相关(分别为χ2=5.827、6.959,P<0.05)。胆囊癌组织中HSP90阳性表达率明显高于正常胆囊组织HSP90阳性表达率82.86%(58/70)比25.71%(9/35),χ2=32.993,P<0.01],差异有统计学意义,HSP90表达水平与胆囊癌患者组织学分化程度、淋巴结转移、TNM分期明显相关(分别为χ2=6.744、7.392、6.110,P<0.05)。结论胆囊癌组织中EIF5A2与HSP90表达上调,EIF5A2和HSP90可能在一定程度上参与胆囊癌的发生发展过程。

Abstract:Objective To investigate the expression of eukaryotic translation initiation factor 5A2(EIF5A2)and heat shock protein 90(HSP90)in gallbladder cancer(GBC)tissues and their clinical significance.Methods Totally 70 cases of tissues pathologically confirmed from March 2014 to October 2020 on Shanxi People′s Hospital were collected,and 35 cases of normal gallbladder tissues resected due to gallstone were enrolled as control group.EIF5A2 and HSP90 expression levels in these tissues were examined by envision immunohistochemical method.The statistical analysis of chi-square test was carried out in combination with GBC clinical pathological features.Results The expression rate of EIF5A2 in GBC tissues was higher than that in normal gallbladder tissues72.86%(51/70)vs.8.57%(3/35),χ2=38.603,P<0.01].The expression of EIF5A2 was associated with lymph node metastasis,and tumor node metastasis(TNM)stage of GBC(respectively,χ2=5.827,6.959,P<0.05).The expression rate of HSP90 in GBC tissues was higher than that in normal gallbladder tissues82.86%(58/70)vs.25.71%(9/35),χ2=32.993,P<0.01].The expression of HSP90 was associated with lymph node metastasis,histology grade and TNM stage of GBC(respectively,χ2=6.744,7.392,6.110,P<0.05).Conclusion EIF5A2 and HSP90 protein expression increased in GBC tissues,which may play a certain degree role in the occurrence and development of GBC.
