当归补血汤对糖尿病肾病大鼠肾损伤的保护作用研究

目的:初步探讨当归补血汤对气血双虚糖尿病肾病大鼠的肾组织保护作用。方法:采用高糖高脂饲料及腹腔注射链脲佐菌素(STZ),结合环磷酰胺和放血法建立气血双虚糖尿病肾病大鼠模型,检测成功后模型大鼠按照随机分组的方法,分为模型对照组、当归补血汤(DBD)高剂量组、DBD低剂量组、贝那普利(BN)组,每组10只,雌雄各半,另选10只大鼠设为正常对照组。灌胃给予当归补血汤前检测各组受试动物的血液学指标,灌胃给予当归补血汤8周后称量各组受试动物的体质量,计算肾体比,检测血糖、尿蛋白、血肌酐、尿素氮、血液学等指标,并对肾脏组织的病理形态学改变进行观察。结果:各组受试动物的血液学指标与给药前相比有显著提高;与模型对照组比较,DBD受试动物的体质量显著增加,肾体比显著减小,血糖、尿蛋白、血肌酐、尿素氮等指标以及肾脏组织的病理形态学改变有明显改善。结论:当归补血汤对于糖尿病肾病模型大鼠具有补气补血,控制肾脏肥大,改善肾组织损伤的作用。

Experimental Study on the Renal Protection of Danggui Buxue Decoction in Rats with Diabetic Nephropathy of Defiency of Qi and Blood

Objective:To preliminarily explore the renal protection of Danggui Buxue Decoction in rats with diabetic nephropathy(DN)of deficiency of qi and blood.Methods:DN model of deficiency of qi and blood was established by high fat diet and streptozotocin(STZ)intraperitoneal injection combined with Cyclophosphamide and bloodletting method.The successful models were randomly divided into the model control,the Danggui Buxue Decoction groups(DBD groups)of high-dose and low-dose,and the Benazepril group(BN group),with 10 rats in each group.Another normal 10 rats were taken as the normal control.Hematology indexes of each group were measured before the treatment of Danggui Buxue Decoction.After 8 weeks of the treatment,body mass,kidney weight/body weight,blood glucose(FBG),urine protein,serum creatinine(Cr)and urea nitrogen(BUN)were measured.Meanwhile,the pathological morphological changes of renal tissue were observed in the groups.Results:Compared to those before the treatment,hematology indexes were significantly increased after the treatment in the groups.Compared to those in the model control,the body mass was increased significantly,kidney weight/body weight was decreased,FBG,urine protein,serum Cr,BUN and the pathological morphological changes of renal tissue had improved significantly in the rats.Conclusion:Danggui Buxue Decoction can invigorate qi and blood,control kidney hypertrophy and improve renal tissue damage.
