摘    要:目的 分离纯化SD大鼠胰腺导管上皮细胞,为将其转分化为胰岛细胞提供细胞来源。方法 用酶消化法培养SD大鼠胰腺导管上皮细胞,观察细胞增殖动态,用CK-19进行免疫组化鉴定。结果 胰腺导管上皮细胞可在体外有效扩增,有效去除成纤维细胞是扩增的关键。结论 低血清培养和反复贴壁法可获得较纯的胰腺导管上皮细胞。

Abstract:Objective To isolate and purify pancreatic ductal epithelial cells for further differentiation towards pancreatic islet cells. Methods The neonate rat pancreas were digested with collagenase and pancreatic ductal epithelial cells were obtained. The cells were observed for their proliferation and were identified with CK 19. Results The pancreatic ductal epithelial cells proliferated better if the fibroblasts were cleared efficiently. Conclusion Low concentration of serum cultivation and adherence repeatedly could let us obtain purified pancreatic ductal epithelial cells.
