Simultaneous Determination of Six Compounds in Rat Plasma by Ultra‑Performance Liquid Chromatography with Tandem Mass Spectrometry: Application in the Pharmacokinetic Study of Qing Gan‑Shu Yu‑Fang

A rapid and high selective ultra?performance liquid chromatography (UPLC) with tandem mass spectrometry method for simultaneous determination of six compounds including albiflorin, paeoniflorin, picroside I, picroside II, saikosaponin A, and saikosaponin D in rat plasma was developed and validated using butyl p-hydroxybenzoate as an internal standard. One-step direct protein precipitation with acetonitrile was used to extract the compounds from the rat plasma samples. Chromatographic separation was achieved using an ACQUITY UPLC BEH C18 column (100 mm × 2.1 mm, 1.7 µm) at a flow rate of 0.4 mL/min, using gradient mode containing 0.1% formic acid in water and acetonitrile were used as the Mobile phase A and B. Electrospray ionization in negative ion mode and multiple reaction monitoring were used to identify and quantify active components. Calibration curves showed good linearity (R2 > 0.9908) over a wide concentration range for all compounds. The intra- and interday precision (relative standard deviation) ranged 2.4%–7.0% and 2.6%–8.0%, respectively. The accuracy (relative error) was from ?13.0% to 13.2% at all quality control levels. The recovery ranged from 81.1% to 92.5%. The validated method was successfully applied to pharmacokinetic study in rats after oral administration of Qing Gan?Shu Yu?Fang. The results show that one can draw a conclusion that these six active ingredients can be quickly absorbed and play a pharmacodynamic role rapidly in vivo.     

miR-138介导ERK和AKT对NSCLC细胞株A549增殖和迁移的作用机制研究

目的探讨miR-138介导ERK和AKT对非小细胞肺癌(NSCLC)细胞株A549增殖和迁移的作用机制。方法选取NSCLC细胞株A549分成Az组、Am组和Ay组,Az组A549细胞转染空载体,Am组转染miR-138-mimics,Ay组转染miR-138-inhibitions,采用RT-PCR法检测miR-138、ERK和AKT mRNA水平变化,Westernblot法检测ERK和AKT蛋白,分别用Transwell法和MTT法检测A459细胞的迁移和增殖情况,流式细胞仪检测A549细胞凋亡程度。结果 RT-PCR检测三组miR-138、ERK和AKT mRNA水平变化:Ay组ERK和AKT mRNA水平变化最高,Az组其次,Am组最低; Am组miR-138水平变化最高,Az组其次,Ay组最低(均P <0. 05)。Westernblot法检测三组ERK和AKT蛋白表达:Ay组ERK和AKT蛋白表达最高,Az组其次,Am组最低(均P <0. 05)。流式细胞仪检测三组A549细胞凋亡程度:Am组A549细胞凋亡最多,Az组其次,Ay组最低,Am组A549细胞凋亡数量较Ay组多,说明A549细胞凋亡随miR-138水平增加而上升(均P <0. 05)。Transwell检测三组A549细胞迁移能力:Ay组癌细胞迁移能力强,显微镜下细胞数量多,Am组癌细胞数量较Az组少,迁移能力较弱(均P <0. 05)。MTT检测三组A549细胞增殖能力:Ay组细胞增殖能力最高,Az组其次,Am组最弱(均P <0. 05)。结论 miR-138在NSCLC细胞株A549的迁移和增殖中发挥重要作用,其过表达可调控ERK和AKT信号,抑制细胞迁移和增殖,促进细胞凋亡。     

Musashi-2在宫颈癌中高表达并促进宫颈癌细胞恶性生物学行为

目的探索Musashi-2 (MSI2)在宫颈癌中的表达水平及其对宫颈癌细胞的作用,为宫颈癌的MSI2靶向治疗提供新的思路和理论基础。方法采用免疫组化法检测53例宫颈癌及20例非恶性子宫病变宫颈组织(对照组)中MSI2相对表达水平,分析宫颈癌中MSI2蛋白表达与临床病理参数(肿瘤分级、分期、患者年龄、淋巴结转移)的关系。采用小RNA干扰(siRNA)技术敲除He La细胞中MSI2建立实验组和对照组,分别采用聚合酶链式反应和蛋白质印迹技术检测宫颈癌细胞中MSI2在mRNA和蛋白质水平的相对表达量。采用CCK-8法检测细胞增殖能力的变化,划痕试验检测细胞迁移能力,Transwell试验检测细胞侵袭能力。结果宫颈癌组织中MSI2蛋白阳性表达率67.92%明显高于对照组的25.00%,差异有统计学意义(P0.05)。MSI2高表达与宫颈癌分期、分级和淋巴结转移正相关(P0.05),与患者年龄无关。MSI敲除后He La细胞增殖、侵袭和迁移能力均有所降低,差异有统计学意义(P0.05)。结论 MSI2蛋白在宫颈癌中通过促进癌细胞增殖、侵袭和迁移导致宫颈癌恶性程度增加,研究为宫颈癌的MSI2靶向治疗提供了新的思路和理论基础。
