豚鼠耳蜗微血管内皮细胞体外通透性的研究   总被引:1,自引:0,他引:1  
目的 研究豚鼠耳蜗微血管内皮细胞的体外通透性特征。方法 在成功建立豚鼠耳蜗微血管内皮细胞体外通透性模型的基础上 ,研究该体外模型跨细胞电阻及对12 5I 牛血清白蛋白的通透性。结果 ①耳蜗微血管内皮细胞在培养增殖过程中其跨细胞电阻呈动态变化过程 ,以 5× 10 4/cm2 的细胞密度接种时 ,7d左右电阻到达峰值 ,其跨细胞电阻峰值大小为 (118 9± 18 5 )Ω/cm2 ;②体外模型中加入12 5I 牛血清白蛋白后 ,其通透性呈上升期抛物线型曲线 ,90min内几乎呈直线。结论 跨细胞电阻及对12 5I 牛血清白蛋白变化曲线能反映体外耳蜗微血管内皮细胞的通透性特征  相似文献

The thiadiazinone derivative [+]-EMD 60263 ((+)-5-(1-(α-ethylimino-3,4-dimethoxybenzyl)-1,2,3,4- tetrahydroquinoline-6-yl)-6-methyl-3,6-dihydro-2H-1,3,4 -thiadiazine-2-on) is a Ca2+-sensitizing agent with only minor phosphodiesterase inhibitory activity. Our aim was to characterize the inotropic and electrophysiological effects of [+]-EMD 60263 and its enantiomer [-]-EMD 60264 in several cardiac muscle preparations. The Ca2+-sensitizing activity resided in the [+]-enantiomer only. [+]-EMD 60263 (3 μM) shifted the EC50 of Ca2+ for contractile activation of skinned fibers of pig heart from 2.41 μM to 0.73 μM, whereas [-]-EMD 60264 (30 μM) was ineffective. In Langendorff-perfused guinea pig hearts, [+]-EMD 60263 and [-]-EMD 60264 induced concentration-dependent positive and negative inotropic effects, respectively; both enantiomers reduced spontaneous heart rate but did not influence perfusion pressure. The maximum increase in force of human atrial trabeculae was 35 % of pre-drug control with [+]-EMD 60263 in comparison to 113 % with forskolin. In guinea-pig papillary muscles, [+]-EMD 60263 and [-]-EMD 60264 had opposite inotropic responses, however, both agents similarly prolonged action potential duration. Both enantiomers concentration-dependently blocked the rapidly activating component IKr of the delayed rectifier in guinea-pig myocytes. The block saturated at potentials positive to +30 mV, closely resembling the effects of the antiarrhythmic agent E-4031 which had been originally used to define IKr. It is concluded, that the positive inotropic action of [+]-EMD 60263 can be explained by prevalence of the Ca2+-sensitizing effect. The accompanying prolongation in action potential duration is caused by block of the IKr component of the delayed rectifier. While the inotropic effects are stereoselective, most of the electrophysiological actions are clearly independent of sterical configuration. The combination of Ca2+-sensitizing with class-III antiarrhythmic action may provide an interesting pharmacological profile of potential therapeutic use. Background Interleukin-8 (IL-8) hus been shown to be a chemotactic factor for netitrophils, T-lymphocytes and eosinophils. Repeated intranasal administration of IL-8 enhances bronchial responsiveness to inhaled histamine in guinea-pigs. Neuropeptides which arc released trotn C-fibre nerve-endings have been postulated to induce bronchial hyperresponsiveness through neurogenic inflammation. Objective This study was conducted to examine whether sensory neuropeptides are involved in the IL-8-induced bronchial hyperresponsiveness. Methods IL-8 at a dose of 5μg/kg was administered intranasally to guinea-pigs twice a week for 3 weeks. One day after the last administration, animals were anesthetized and artificially ventilaled through tracheal cannula, and lateral pressure at the tracheal cannula (Pao) was measured as an overall index of airway responses lo increasing concentrations of inhaled histamine (25, 50, 100, and 200 μg/mL). A NKI and NK2 dual antagonist FK224(10mg/kg), a selective NK1 antgonist FK888 (10mg/kg) or vehicle was intravenously administered 10min before measurement of bronchial responsiveness. Result The IL-8 treatment significantly enhanced bronchial responsiveness to histamine (ANOVA P < 0.01). FK224 or FK888 did not alter the IL-8-induced bronchial hyperresponsiveness. Conclusion We conclude that repeated intranasal administratioti of IL-8 causes bronchial hyperresponsiveness (BHR) and that neuropeptides such as neurokinin A and substance P do not directly contribute to the development of BHR induced by IL-8.  相似文献

自身免疫性感音神经性聋豚鼠的子代内耳生理功能研究   总被引:4,自引:4,他引:0  
目的 :观察自身免疫性感音神经性聋 (ASHL)母豚鼠所产子代内耳生理功能的变化 ,探讨针对内耳的自身免疫因素对子代内耳生理功能的影响及其改变特点。方法 :同种内耳抗原 (CIEAg)持续免疫孕豚鼠 ,采用耳蜗电图 (记录cAP、CM )和眼震电图仪 (记录自发性眼震和冷热空气试验 )测试母鼠和子鼠的听觉和前庭功能 ,并检测血清特异性体液免疫反应。结果 :ASHL模型母豚鼠所产子鼠血清中发现有特异性抗体水平升高 ,部分 (3 /9)出现听觉损伤。非ASHL母鼠和对照组母鼠所产子代未见明显异常。结论 :ASHL雌鼠所产子代可出现感音神经性聋 ,其内耳损伤和功能障碍极可能与针对内耳组织的自身免疫反应 (尤其是体液免疫 )有关 ,从而提示内耳自身免疫因素可能为部分先天性非遗传性感音神经性聋的病因之一。  相似文献

Summary Changes in regional cerebral blood flow in anaesthetized pigs with an induced focal cerebral contusion were studied before and after two grades of hyperventilation. A reduction in arterial tension of CO2 with 0.70mmHg and a further reduction of 0.55mmHg did not change the CO2 reactivity. Reactivity in both injured and macroscopically normal regions was the same, revealing an average of 39.3% flow change per kPa change in CO2 tension. Regions with low flow after the contusion had an equally big reduction apparently leading to hypoxia because global metabolic rate was unchanged.  相似文献

本研究以豚鼠皮层听觉诱发电位反应阈为指标,采用自体或群体对照方法,观察0.1Hz极低频磁场对豚鼠听力的影响。结果表明,豚鼠经50mT(毫特)、4h,100mT、48h及10mT、4周暴露后,听阈降低;50mT、48h,20mT、4周暴露后听阈无明显变化。提示0.1Hz极低频磁场对豚鼠听敏度无不利的影响,在某些暴露条件下,听敏度尚有改善。  相似文献

目的 研究卡介苗对哮喘豚鼠的防治作用。方法 24只豚鼠随机分为阳性对照组、地塞米松组、卡介苗组及阴性对照组,每组6只,除阴性对照组用生理盐水外,其余三组每只腹腔内注射鸡卵蛋白100mg,2周后用1%鸡卵蛋白激发致敏,每次30min,每日1次,持续3d,地塞米松组每次激发前肌注地塞米松2mg/kg,卡介苗组每次激发前肌往卡介苗30mg/kg,阳性对照组每次激发前30min肌注生理盐水2ml,最后用乌拉坦麻醉,检验动脉血气分析参数,嗜酸粒细胞(EOS)计数等参数,所有参数用t检验或卡方检验分析。结果 阳性对照组与其它三组比较,在豚鼠的发病程度、动脉血气分析、外周血EOS计数、支气管肺泡灌洗液中EOS计数、支气管病理切片每20个高倍视野EOS计数均有明显差异(P<0.05或P<0.01)。结论 卡介苗能够抑制动物模型中的哮喘反应。  相似文献

Summary The influence of locomotor activity upon neurons in the lateral vestibular nucleus was investigated in precollicularly-postmamillary decerebrate guinea pigs. Out of 95 recorded neurons, 24 were identified as vestibulospinal and 71 had no descending projections. Locomotor activity occurred either spontaneously or was prompted by electrical stimulation of the mesencephalic locomotor region. Natural vestibular stimulation was supplied by tilting the animal about its longitudinal axis. Locomotor rhythmic limb muscle activity was accompanied by an increase in the firing frequency in the vast majority of investigated neurons. The increase in frequency was observed at the beginning of ipsilateral forelimb extensor muscle activity. Only in a few non-vestibulospinal neurons was the spontaneous activity depressed during locomotion. An increase in evoked responses was observed in almost all vestibulospinal neurons and in two thirds of the neurons without descending projections. A decrease in evoked responses was observed in one quarter of non-vestibulospinal neurons. During locomotion, the mean and maximal frequencies of evoked neuronal impulse activity changed, but the phase lag of these changes was not altered significantly. The results suggest an enhancement of vestibulospinal influences during locomotion, thus providing a high level of tonus in antigravitational muscles. This is interpreted as a mechanism to ensure that equilibrium is maintained during motion in different gaits and postures.  相似文献

Objective: To explore the protective effects of earplug and barrel on auditory organs of guinea pigs exposed to experimental blast underpressure (BUP). Methods : The hearing thresholds of the guinea pigs were assessed with auditory brainstem responses ( ABR). The traumatic levels of tympanic membrane and ossicular chain were observed under stereo-microscope. The rate of outer hair cells (OHCs) loss was analyzed using a light microscope. The changes of guinea pigs protected with barrel and earplug were compared with those of the control group without any protection. Results: An important ABR threshold shift of the guinea pigs without any protection was detected from 8h to 14d after being exposed to BUP with a peak ranging from -64.5 kPa to -69.3 kPa (P<0.01). The rate of perforation of tympanic membrane reached 87.5 % and that of total OHCs loss was 19.46%±5.38% at 14d after exposure. The guinea pigs protected with barrel and earplug had lower ABR threshold and total OHCs loss rate compared with the animals without any protection (P<0.01). All of the tympanic membrane and ossicular chain of the protected animals maintained their integrities. Meanwhile, the guinea pigs protected with the barrel had lower ABR threshold and total OHCs loss rate than those with earplug (P<0.01). Conclusions: The earplug and barrel have protective effects against BUP-induced trauma on auditory organs of the guinea pigs and the protective effects of barrel are better than those of earplug.  相似文献

用低蛋白饮食方法建立豚鼠胆色素结石模型,共设对照、致石、维生素C修复、丹参修复和对照修复等5组,规定时间内处死动物,用放射免疫、固相酶联免疫、生物化学等方法检测肝细胞内环—磷酸腺甙(cAMP)、环—磷酸鸟嘌呤(cGMP)、钙调素(CaM)、钙,三磷酸腺甙酶(Ca2+-ATPase)、磷酸化酶a等水平。致石组豚鼠肝脏细胞内cAMP和磷酸化酶a升高,而cGMP,CaM和Ca2+-ATPase下降,表明肝细胞钙稳态呈失调状态。维生素C和丹参可调整肝细胞的上述改变,说明维生素C和丹参具有维持肝细胞钙稳态的作用。  相似文献
