BACKGROUND: Microbial intestinal colonization in early in life is regarded to play a major role for the maturation of the immune system. Application of non-pathogenic probiotic bacteria during early infancy might protect from allergic disorders but underlying mechanisms have not been analysed so far. OBJECTIVE: The aim of the current study was to investigate the immune effects of oral application of probiotic bacteria on allergen-induced sensitization and development of airway inflammation and airway hyper-reactivity, cardinal features of bronchial asthma. METHODS: Newborn Balb/c mice received orally 10(9) CFU every second day either Lactobacillus rhamnosus GG or Bifidobacterium lactis (Bb-12) starting from birth for consecutive 8 weeks, during systemic sensitization (six intraperitoneal injections, days 29-40) and airway challenge (days 54-56) with ovalbumin. RESULTS: The administration of either Bb-12 or LGG suppressed all aspects of the asthmatic phenotype: airway reactivity, antigen-specific immunoglobulin E production and pulmonary eosinophilia (mean: 137 vs. 17 and 13 cellsx10(3)/mL, respectively). Antigen-specific recall proliferation by spleen cells and T-helper type 2 cytokine production (IL-4, IL-5 and IL-10) by mesenteric lymph node cells also showed significant reduction, while TGF production remained unchanged. Oral LGG administration particularly suppressed allergen-induced proliferative responses and was associated with an increase in numbers of TGF-beta-secreting CD4+/CD3+ T cells in mesenteric lymph nodes (6.5, 16.7%) as well as nearly 2-fold up-regulation of Foxp3-expressing cells in peribronchial lymph nodes. CONCLUSIONS: Neonatal application of probiotic bacteria inhibits subsequent allergic sensitization and airway disease in a murine model of asthma by induction of T regulatory cells associated with increased TGF-beta production. 相似文献

目的:观察从健康核潜艇艇员正常粪便中分离出的双歧杆菌QJ405和乳酸杆菌QJ405的生物学特征,判定其是否符合益生菌要求。方法:分析细菌的形态学和生理生化特征。采用数值分类法(API20A)的化学分类法(气相色谱分析)进行种属鉴定。抗生素敏感试验采用ATB法。结果:两株菌分别鉴定为长双杆菌和唾液乳酸杆菌唾液亚种,两菌代谢发酵碳水化合物均可产生大量乙酸、乳酸,对多种抗生素表现敏感。结论:双歧杆菌QJ405和乳酸杆菌QJ405的来源和部分生物活性符合益生菌的要求。 相似文献

Background and aim: The present study was conducted to determine the abilities of the living Lactobacillus plantarum MON03 (LP) cells, Tunisian montmorillonite clay and their composites to accumulate Zearalenone (ZEA) from a liquid medium and elucidate the preventive effect of their composite in ZEA-contaminated balb/c mice showing immunotoxicity disorders.Materials and methods: In the in vitro study, LP (2 × 109 CFU/mL), TM (0.5 mg) and LP+TM were incubated with 50 µg mL?1 ZEA for 0, 12 and 24 h. For the in vivo study, the composite MT+LP was evaluated also for possible protection regarding ZEA-immunotoxicity in Balb/c mice as a sensitive model.Results: Results indicated that TM and LP+TM had a high capacity of adsorbing ZEA 87.2 ± 2.1 and 94.2 ± 2.1%, respectively. However, LP alone able to remove only 78% after 24 h of incubation. The quantity of adsorbed ZEA by LP, TM and LP+TM were 39, 43,5 and 47 µg mL?1 of PBS, respectively. The in vivo results indicated that mice orally exposed to ZEA- (40 mg/kg bw) for 2 weeks showed severe immunotoxicity typical of fusarotoxicosis regarding thymocytes and splenocytes cell viability count, IFN-γ, IL-12, TNF-α production and B-cell activation. Mice treated with LP and TM alone, and LP+MT in combination with ZEA were comparable to the control.Conclusion: Both LP and TM are safe by themselves and their composite succeeded to exert a potential prevention by counteracting ZEA-immunotoxicity and can be implicated in the biotechnology of ZEA removal in human food and animal feed. 相似文献

Previous in vitro and in vivo studies have suggested that lactobacilli can exert antiproliferative effects on the gastrointestinal epithelium. However, their role in affecting the cellular proliferative mechanisms is not completely clear. The aim of this study was to investigate the effects of increasing concentrations of Lactobacillus rhamnosus strain GG (L. GG) homogenate on cell growth and proliferation (by MTT, [3H]-thymidine incorporation and polyamine biosynthesis) in neoplasms originating from different gastrointestinal tracts. Thus, HGC-27 human gastric cancer cells and DLD-1 human colonic adenocarcinoma cells were evaluated. Besides, in order to verify which bacterial fraction was involved in the antiproliferative effects, the cytoplasm and cell wall extracts were tested separately. Gastric HGC-27 and colonic DLD-1 cells showed significant differences in their proliferative behavior, in particular in their polyamine profile and biosynthesis. Notwithstanding, one and the other proved to be sensitive to the growth inhibition by the highest concentrations of bacterial homogenate. Both HGC-27 and DLD-1 cells were resistant to the bacterial cell wall fractions, whereas increasing cytoplasm fraction concentrations induced an evident antiproliferative effect. These data suggest that cytoplasm extracts could be the responsible for L. GG action on proliferation in these two cell lines from gastric and colonic neoplasms. 相似文献

目的:初步研究乳杆菌A-2代谢产物(LSPM1和LSPM2)对人舌鳞癌CAL-27细胞的增殖抑制及诱导细胞凋亡.方法:乳杆菌A-2通过降解制备相应的代谢产物LSPM1和LSPM2;采用MTT法检测不同浓度的LSPM1和LSPM2(1.875mg/mL、3.75mg/mL、7.5mg/mL、15mg/mL和30mg/mL)对CAL-27细胞的体外增殖抑制作用;荧光显微镜下观察CAL-27细胞形态变化;采用流式细胞仪和DNA琼脂凝胶电泳法检测细胞凋亡情况.结果:不同浓度的乳杆菌A-2代谢产物LSPM1和LSPM2可以抑制CAL-27细胞的增殖,呈现明显的浓度依赖性;LPSM1可明显诱导CAL-27细胞凋亡,呈时间依赖性.结论:乳杆菌A-2代谢产物可以在体外抑制人舌鳞癌CAL-27细胞生长增殖,并诱导CAL-27细胞凋亡. 相似文献

目的 比较以甲硝唑、雌激素和乳酸杆菌制剂局部应用治疗老年性阴道炎的疗效和复发率.方法 以初次患老年性阴道炎的病人为研究对象,分别采用阴道内甲硝唑200mg(Ⅰ组),己烯雌酚0.25mg(U组)和乳酸杆菌制剂0.25g(Ⅲ组).观察外阴瘙痒、白带增多症状和阴道黏膜充血和点状出血的体征,判断有效率和一月、三月复发率.结果 外阴瘙痒症状的改善第Ⅲ组较其他两组明显,白带增多症状改善Ⅱ、Ⅲ组较第Ⅰ组明显,体征的改变第Ⅲ组较第Ⅰ、Ⅱ组显著;总有效率第Ⅲ组达90%,明显高于其他两组;一月、三月复发率明显低于其他两组.结论 乳酸杆菌制剂用于老年性阴道炎的治疗效果较好,复发率较低. 相似文献

目的调查妊娠期细菌性阴道病(BV)的发病情况,观察定君生胶囊治疗细菌性阴道病的有效性。方法采用珠海市银科生物技术应用研究所生产的细菌性阴道病快速检测卡,对不同孕周的1500名孕妇进行BV筛查,确定妊娠期细菌性阴道病的发病率。对确诊为BV的孕妇266例,随机分为两组。定君生组:136例,给予定君生胶囊(乳杆菌活菌胶囊)阴道给药治疗;孚舒达组:130例,给予孚舒达栓(复方甲硝唑栓)阴道给药治疗。两组均连续用药10d,停药7d后复查。结果妊娠期BV的检出率为17.73%。定君生胶囊治疗BV的总有效率为92.65%,孚舒达栓治疗BV的总有效率为91.53%,二者差异无显著性(P>0.5)。结论妊娠期BV的发病率较高。定君生胶囊能有效改善阴道内生态环境,治疗妊娠期细菌性阴道病安全、有效,易为孕妇所接受。 相似文献
