目的:建立紫杉药材中紫杉烷类和双黄酮类成分的薄层色谱鉴别法。方法:以紫杉宁为对照品,石油醚-乙酸乙酯-丙酮-甲醇(8∶2∶2∶1)为展开剂系统,用硅胶G板鉴别紫杉烷类成分;以金松双黄酮和银杏双黄酮为对照品,石油醚-乙酸乙酯-甲醇(4∶1∶1)为展开剂系统,用聚酰胺薄膜鉴别双黄酮类成分。结果:本方法斑点清晰,重复性好,并可将紫杉区别于一些松、杉、柏类植物的枝叶。结论:本方法简便,快速,可用于紫杉药材的质量控制。  相似文献

目的了解我院紫杉醇治疗肺癌的不良反应发生情况。方法对2004年7月至2009年7月我院应用紫杉醇化疗药治疗肺癌患者96例所发生的不良反应进行回顾性分析。结果本组紫杉醇的主要不良反应是骨髓抑制(白细胞减少57例,血小板减少16例,红细胞减少22例)、神经毒性(35例)、消化道反应(恶心呕吐56例,食欲减退43例,腹泻9例)、过敏反应(19例)、神经毒陛(35例)、心脏章陛(9例)、肝功能异常(6例)等。结论只要做好各种防治措施并掌握好合适的剂量,紫杉醇还是一种较为安全的广谱抗癌药,有着广阔的应用前景。  相似文献

Increased glycation of macromolecules via the reactive dicarbonyl and α‐oxoaldehyde methylglyoxal (MG) has shown an association with diabetes and its complications. In the present study, the protective effects of sciadopitysin against MG‐induced oxidative cell damage were investigated in the insulin‐producing pancreatic β‐cell line, RIN‐m5F cells. When exposed to MG for 48 hours, RIN‐m5F cells experienced significant loss of viability and impaired insulin secretion; however, treatment with sciadopitysin protected RIN‐m5F cells against MG‐induced cell death and decreased insulin secretion. Treatment of RIN‐m5F cells with sciadopitysin prevented MG‐induced production of interleukin‐1β, intracellular reactive oxygen species and cardiolipin peroxidation. Furthermore, sciadopitysin increased adenosine monophosphate‐activated protein kinase phosphorylation of RIN‐m5F cells. Treatment of cells with sciadopitysin increased the activity of glyoxalase I and decreased the levels of MG‐protein adducts, indicating that sciadopitysin protects against MG‐induced protein glycation by increasing MG detoxification. Taken together, the results indicated the potential utility of sciadopitysin as an intervention against MG‐induced cell damage in pancreatic β ‐ cells.  相似文献

目的:建立以高效液相色谱法测定云南红豆杉枝叶中金松双黄酮含量的方法。方法:色谱柱为Agilent Zorbax SB-C_(18) (250 mm×4.6 mm,5μm),流动相为甲醇-水(80:20),流速为1.0 mL·min~(-1),检测波长为272 nm,柱温为40℃结果:金松双黄酮的进样量在0.88~4.40μg·mL~(-1)范围内与峰面积积分值呈良好线性关系(r=0.9999);平均回收率为95.40%,RSD= 3.5%(n=6)。结论:本方法准确、可靠、稳定,可用于云南红豆杉的质量控制。  相似文献

Age-related osteoblast dysfunction is the main cause of age-related bone loss in both men and women. In the present study, the effect of sciadopitysin, a type of biflavonoids, on osteoblast function was investigated in osteoblastic MC3T3-E1 cells. Sciadopitysin caused a significant elevation of alkaline phosphatase activity, collagen synthesis, osteocalcin production, mineralization, and glutathione content in the cells (P < 0.05). Sciadopitysin also decreased the production of tumor necrosis factor-a (TNF-α) induced by antimycin A, a mitochondrial electron transport inhibitor. We investigated the protective effects of sciadopitysin on antimycin A-induced toxicity in osteoblastic MC3T3-E1 cells. Exposure of MC3T3-E1 cells to antimycin A caused a significant reduction in osteoblast dysfunction. However, pretreatment with sciadopitysin prior to antimycin A exposure significantly reduced antimycin A-induced cell damage by preventing mitochondrial membrane potential dissipation, adenosine triphosphate (ATP) loss, reactive oxygen species (ROS) release, and nitrotyrosine increase, suggesting that sciadopitysin may be useful for protecting mitochondria against a burst of oxidative stress. Moreover, sciadopitysin increased phosphorylation of cAMP-response element-binding protein (CREB) inhibited by antimycin A. Our results demonstrate that sciadopitysin may reduce or prevent osteoblasts degeneration.  相似文献
