刺五加叶皂甙单体Sb对培养大鼠心肌细胞动作电位的影响   总被引:1,自引:0,他引:1  
本研究中所用的刺五加叶皂甙单体Sb(50、200μg/mL),使培养的wistar大鼠心肌细胞动作电位的波幅、波宽、阈电位、最大舒张电位、超射、最大除极速度及复极(10%、50%、90%)水平的动作电位波宽一致减小。Ca2+80μg/mL能使之反转,Sb作用与尼莫地平作用相似。上述结果表明Sb具有钙通道阻滞作用。  相似文献

目的 建立喜炎平注射液中17种重金属及有害元素的含量测定方法。方法 样品经消解后采用电感耦合等离子体质谱(ICP-MS)法测定,射频功率1 550 W;蠕动泵转速0.3 r/s;等离子气体积流量15 L/min;辅助气体积流量0.2 L/min;载气体积流量1 L/min;采样深度10 mm,重复次数为3次。结果 该方法下Li、Al、V、Cr、Fe、Co、Ni、Cu、As、Ag、Cd、Sn、Sb、Ba、Hg、Pb、Bi的检测限分别为9.584、49.858、0.504、3.016、51.209、0.142、1.116、0.675、0.924、1.421、0.403、2.770、0.711、3.584、0.590、0.411、0.169 ng/mL,定量限分别为28.933、151.085、1.528、9.139、155.179、0.429、3.381、2.046、2.799、4.312、1.220、8.394、2.155、10.861、1.965、1.244、0.513 ng/mL,各元素在一定质量浓度范围内,响应值与质量浓度的线性关系良好(r>0.999 1),进样精密度RSD在0.8%~3.8%(n=12),重复性RSD在0.7%~2.0%(n=6),17种元素的加样回收率在95.7%~104.8%。20批大生产样品检测结果:Ag为未检出,Li和V为检出但低于定量限,Al、Ni、Cu、As、Ba、Hg、Pb最大质量浓度分别为0.685、0.013、0.007、0.006、0.208、0.070、0.027 μg/mL,Cr、Fe、Co、Cd、Sn、Sb、Bi为未检出或检出但低于定量限。结论 方法的专属性、线性、灵敏度、精密度、准确度、耐用性等均良好,可用于喜炎平注射液中Li、Al、V、Cr、Fe、Co、Ni、Cu、As、Ag、Cd、Sn、Sb、Ba、Hg、Pb、Bi 17种元素的含量测定,20批大生产样品中17种元素检测结果均小于限度要求,符合规定。  相似文献

目的:建立电感耦合等离子体质谱法测定酒石酸长春瑞滨中有害元素及催化剂银残留的方法。方法:用微波消解处理样品,电感耦合等离子体质谱内标法测定银、钒、铬、钴、镍、铜、砷、钼、镉、锡、锑、钡、铅、汞的含量。结果:上述14种元素的方法检出限分别为0.003、0.020、0.141、0.069、0.077、0.362、0.031、0.057、0.022、0.078、0.017、0.724、0.095、0.005 ng·mL-1,线性关系良好(r≥0.9988),回收率为86%~110%,RSD小于15%;同时,给出了此方法各元素的不确定度评定程序。结论:本方法采用在线加内标,可有效校正仪器漂移,抑制基体干扰,提高准确率,多元素测定便捷、准确、灵敏、可靠,可用于酒石酸长春瑞滨中有害元素及催化剂银残留的测定。  相似文献

Monoclonal antibodies to the 28kDa glutathione S-transferase of Schistosoma bovis have been constructed in mice and used to characterize the epitope(s) potentially implied in the induction of anti-fecundity and anti-egg viability immune responses. Among the MoAbs produced three were particularly studied: Sb4-50 (IgG2a) and Sb4-56 (IgG1) which inhibited Sb28GST activity and Sb4-10 (IgG1) which did not. The use of overlappting peptides covering the entire amino acid sequence of Sb28GST, allowed us to define the linear epitopes recognized by these anti-Sb28GST MoAbs. Amino acid residues 202-211 were recognized by both MoAbs Sb4-50 and Sb4-56 and MoAb Sb4-10 recognized amino acid residues 58-67. Their capacity to inhibit GST activity suggested binding to the active site or to neighbouring regions, which include the C-terminal domain (a.a. 190-211) of the protein. When passively transfered into BALB/c mice MoAbs induced a significant reduction in egg hatching and an increase in immature eggs. Effects on worm burdens were, however, variable and no clear-cut association between the inhibition of enzyme activity and anti-fecundity or anti-viability activities was recorded. Our data indicate that beside the anti-fecundity and anti-viability immunity related to the impairment of GST activity, immune response to epitopes located in other regions of the molecule also contribute to the reduction of egg viability .  相似文献

[目的]测定并分析评价烟台市部分海域不同种类海产品中Sb含量水平。[方法]利用氢化物原子荧光光谱法测定海产品中Sb含量,并采用方差分析及重金属单因子污染指数法对不同种类海产品中Sb含量状况进行分析。[结果]15种海产品Sb平均含量为0.006 3mg/kg,其中鱼类为0.004±0.000 4mg/kg,软体动物为0.005±0.000 3mg/kg,甲壳类为0.007±0.000 4mg/kg,贝类为0.009±0.001 1mg/kg,贝类海产品Sb含量显著高于鱼类、软体动物及甲壳类(P<0.05),所有海产品单因子污染指数均小于0.2,其中,蛏子的Sb单因子污染指数最高(0.030 0)。[结论]烟台市黄渤海海域所采集的15种海产品均检出重金属Sb,但Sb浓度处于正常水平,尚未出现Sb污染现象。  相似文献

Background

The assessment of human chemical risks related to occupational or environmental exposure to pollutants requires the use of both accurate exposure indicators and reference values. The objective of this study was to evaluate the blood and urinary levels of various metals and metalloids in a sample of adults aged 20–59 years of the general population of Northern France, a formerly heavily industrialised area that retains some industrial activity.

Methods

A cross-sectional study was conducted between 2008 and 2010, enrolling 2000 residents of Northern France. The quota method was used to guarantee the representativeness of the participants on a sex, age, social category and smoking status basis, according to the census done by the French National Institute of Statistics and Economic Studies. The levels of 14 metals: aluminium (Al), antimony (Sb), total arsenic (As), beryllium (Be), cadmium (Cd), cobalt (Co), chromium (Cr), mercury (Hg), manganese (Mn), nickel (Ni), lead (Pb), thallium (Tl), vanadium (V) and zinc (Zn) were quantified by ICP-MS in urine and blood samples.

Results

A total of 982 men and 1018 women participated, allowing the analysis of 1992 blood and 1910 urine samples. Some metal(loid)s were detected in over 99% of the blood (Cd, Co, Mn, Ni, Pb) and urine (As, Co, Pb, Zn) samples and the remaining metals in 84–99% of the samples, with the exception of blood V (19%), blood Be (57%) and urine Be (58%). Mean blood levels of Pb and Zn were significantly higher in men, and Mn, Co and Cr in women. In urine, mean Pb, Tl and Sb concentrations were significantly higher in men, and Al and Co in women. Current smokers had significantly higher mean levels of blood Cd and Pb and lower blood Co, Mn and Hg. In urine (adjusted on urinary creatinine), the smokers had higher mean levels of Cd, Pb, V and Zn and lower mean levels of As, Co, and Hg. Overall, the mean urinary levels of most metal(loid)s found in the general population of Northern France were higher than those found in the French national survey for the same period except for urinary V. Mean blood lead level was markedly less than that of the French national population.

Conclusion

This first biomonitoring survey of a large number of metal(loid)s in the general population of Northern France provides useful information on exposure levels to toxic elements and highlights the specificity of the regional environment. These data could be used, in complement to the national human biomonitoring reference values, for the interpretation of biomonitoring results.  相似文献

目的 通过电感耦合等离子体质谱(inductively coupled plasma mass spectrometry,ICP-MS)法建立加味左金丸中Cd、Pb、As、Hg、Co、V、Ni、Cu、Li、Sb、Ba、Mo、Sn、Cr、Na、Mg、Al、Ca、Ti、Mn、Fe、Zn、Ga、Se、Sr、Tl共计26种无机元素的测定方法。方法 加味左金丸通过微波消解法处理后,根据相对分子质量的大小选择内标物,其中7Li、23Na、24Mg、27Al、40Ca、48Ti、51V、52Cr、55Mn、56Fe、58Ni、59Co、63Cu、66Zn、70Ga、75As、77Se、86Sr以72Ge作为内标;95Mo、114Cd、118Sn、121Sb137Ba以115In作为内标;202Hg、205Tl、208Pb以209Bi作为内标。对标准品溶液、空白溶液与供试品溶液进行分析,采用标准曲线法进行定量分析。通过ICP-MS法进行测定。结果 26种无机元素线性的相关系数r ≥ 0.999 6,检出限为0.001~1.500μg/L,定量限为0.01~5.00 μg/L,精密度与重复性试验的RSD均小于5%,平均回收率在82.64%~106.44%,RSD均小于5%。对3个厂家的12批样品进行了测定,26种元素的含量差异较大,其中Na、Mg、Ca、Fe 4种元素的含量比较高,均大于500 μg/g,Cd、Pb、As、Hg、Co、Li、Sb、Mo、Sn、Cr、Se、Tl的含量比较低,均小于1 μg/g。由结果可知,人体的常量元素,如Na、Mg、Ca的含量比较高,Cd、Pb、As、Hg等有害元素含量比较低。根据《中国药典》2020年版一部的要求,本品中Cd、Pb、As、Hg与Cu均符合规定。结论 该方法快速、准确,可以用于加味左金丸中无机元素的测定。  相似文献

AIM OF THE STUDY: Leishmaniasis, caused by protozoan from Leishmania genus, is an endemic disease in the tropical and subtropical regions of the world. The chemotherapy to this disease is not always effective and can cause several side effects. Chenopodium ambrosioides L. (Chenopodiaceae) is used by the native people in the treatment of cutaneous ulcers caused by different species of Leishmania. The aim of this study was to investigate the effect of the treatment with a hydroalcoholic crude extract (HCE) from the leaves of Chenopodium ambrosioides on the murine infection with Leishmania amazonensis. MATERIAL AND METHODS: The mice were treated for 4-6 weeks post-infection (p.i.) with HCE (5 mg/kg) or meglumine antimoniate (Sb(v)) (28 mg/kg) either by the oral route, once a day, for 15 days or by five intralesional (IL) injections at intervals of 4 days. The thickness of the infected paws was determined weekly and the parasite load evaluated in the draining lymph nodes (LN), the spleen and in the footpad after 7 weeks of infection. The nitric oxide (NO) production was evaluated in cultures with cells from peritoneum or LN. RESULTS: The IL treatment increased the NO production in the LN and peritoneum cultures and reduced the parasite load from the footpad, spleen and LN. On the other hand, the oral treatment decreased did alter neither the NO production nor the parasite load. CONCLUSIONS: IL HCE treatment was more efficient than the oral HCE treatment since the former was able to control the dissemination of infection. This effect can be due to either a direct leishmanicidal effect of HCE or the improvement in the NO production by HCE-stimulated macrophages. The results could justify the topical use of the Chenopodium ambrosioides' leaves in the treatment of the ulcers caused by Leishmania.  相似文献

目的建立宫瘤消片中铅、镉、砷、汞、锑的原子荧光光谱检测方法。方法采用微波消解法消解宫瘤消片样品后,用原子荧光光谱法测定其中的铅、砷、汞、锑的含量,并对测定方法进行了方法学考察。结果对于所测元素,校准曲线的相关系数r〉0.9992,回收率为90.5%~107.4%,RSD〈7%。结论该方法快速、简便、准确且灵敏度高,为宫瘤消片中重金属含量测定提供了较好的方法,用本方法测定宫瘤消片样品达到满意效果。  相似文献

目的 建立苦参的毛细管电泳指纹数字化图谱(CEFP),考察Jaccard相似度Sb评价苦参CEFP的可行性和特征。方法 采用胶束电动毛细管电泳法,以30mmol·L^-1硼砂+10mmol·L^-1 SDS为背景电解质,运行电压12kV,紫外检测波长205nm,以10个不同产地药材的电泳图谱建立了苦参的对照CEFP。与双定性相似度SF和S′F、双定量相似度C%和P%相比较考察Jaccard相似度参数Sb的可行性和特征。结果 以萘普生为参照物峰,确定15个共有峰,建立了苦参CEFP,并用“中药色谱指纹图谱超信息特征数字化评价系统3.0”软件挖掘苦参CEFP的指纹特征,结合双定性双定量相似度法客观、全面地评价了不同产地苦参的质量。结论 所建立的苦参毛细管电泳数字化指纹图谱具有较好的精密度和重现性,可为苦参质量控制提供新参考,Jaccard相似度Sb参数能够作为评价中药批间相似度一个较好的指标。  相似文献
