A simple method using high-performance liquid chromatography and fluorescence detection is presented for the simultaneous
determination of the antiparasitic agents avermectins (abamectin, ivermectin and doramectin) in bovine liver. Samples were
extracted using acetonitrile and cleaned up on an alumina SPE column and derivatized with trifluoroacetic anhydride. The derivatives
were detected using fluorescence detector set at 365 nm excitation and 475 nm emission wavelengths. As the results, the limit
of quantification for three avermectins was about 1 μg/kg. Inter-assay showed the mean recoveries of avermectins in bovine
liver ranged from 72% to 81%, with RSD less than 9.9%. 相似文献

In this study, linear relationships between response and concentration were used to estimate the detection limit (DL) and quantification limit (QL) for five avermectins: emamectin, abamectin, doramectin, moxidectin, and ivermectin. Estimation of DL and QL was based on the standard deviation of residual and y-intercept of the regression line at low concentrations of avermectins, using the dispersive solid-phase extraction procedure. Avermectin extracts were analyzed using liquid chromatography tandem mass spectrometry. Based on the regression slope, DL and QL were higher at concentrations of 0.3–0.4 μg/kg and 1 μg/kg, respectively, for all avermectin compounds. Linearity assessment was performed by linear regression, which incorporated a regression model, outlier rejection, and evaluation of the assumption with a significant test. For all avermectins, there is a significant correlation between response and concentration in the range 1–15 μg/kg, and the y-intercept passes through origin (zero). 相似文献

目的 :以UV法测定阿维菌素明胶微球 (AVM -GMS)体外释放中阿维菌素 (AVM)含量。方法 :采用双波长法在等吸收点 2 4 5nm、2 74 .5nm处测定吸收度 ,消除干扰物质影响。结果 :吸收度差值与AVM浓度线性关系良好 ,RSD值为 1 94 % ,平均回收率为 95 94 % ;AVM -GMS和AVM GMS(阿维菌素和空白微球混合物 )在一段时间内释放速度被准确测定。结论 :该方法简便易行 ,准确可靠地预测了阿维菌素明胶微球的长效缓释作用 相似文献

以Streptomyces avermitilis ATCC31272和它的突变株为试验菌株进行avermectins发酵培养基的研究,最初筛选到的发酵培养基中必需加入番茄酱和鲜酵母,进一步通过对170多种发酵培养基的筛选实验,选择出一种以淀粉,酵母粉,豆饼粉和无机盐为营养物的培养基,并以它为基础研究了碳源,氮源,无机盐等对avermectins生物合成的影响,经多次培养基的优化试验,突变株Sa-76-9摇瓶发酵效价达3500-4000ug/ml。 相似文献

目的研究阿维菌素原药诱导大鼠肝组织HSP70基因表达的变化,为阿维菌素原药环境污染早期监控及暴露人群的保护提供依据。方法将清洁级Wistar大鼠随机分成3个染毒组和1个对照组,每组12只,雌雄各半,3个染毒组每日经口灌胃不同浓度阿维菌素原药植物油稀释液,最终染毒剂量分别为2.5、1.25、0.625 mg/kg,连续染毒14 d,对照组给予等量花生油。观察大鼠一般状况,测定血清总蛋白(TP),并运用RT-PCR扩增技术观察肝组织HSP70 mRNA表达的变化。结果2.5 mg/kg剂量组雌性大鼠血清TP低于对照组(P<0.05);各染毒组动物肝组织HSP70 mRNA表达呈剂量-效应关系,2.5 mg/kg及1.25 mg/kg剂量组HSP70 mRNA表达明显增加(P<0.05)。结论阿维菌素原药能够敏感地诱导HSP70基因的表达,提示检测HSP70基因表达可作为阿维菌素原药早期监控的指标之一。 相似文献

目的探讨纳洛酮治疗阿维菌素中毒的临床疗效。方法将32例农药阿维菌素中毒患者分为治疗组和对照组,对照组采用常规治疗,清水洗胃,吸氧、补液、利尿,保护肝、肾功能,抗生素及对症支持治疗,治疗组是在常规治疗的基础上加用纳洛酮0.8 mg静脉推注,然后以5%葡萄糖250 mL加纳洛酮1.2 mg持续静脉滴注(滴速2 mL/min)至患者神志转清,然后改为5%葡萄糖250 mL加纳洛酮1.6 mg静脉滴注,每日1次,连用3 d。两组根据治愈率、死亡率、平均清醒时间来判定疗效。结果治疗组总治愈率为93.75%,中毒昏迷患者平均清醒时间为(5.70±2.10)h;对照组治愈率为62.50%,中毒昏迷患者平均清醒时间为(12.44±6.07)h,两组比较差异有统计学意义(P〈0.05)。结论纳络酮可提高抢救成功率及缩短平均清醒时间。 相似文献

目的 设计优化一种化学合成发酵培养基,为阿维菌素产生菌生理生化研究提供基础。方法 在单因素实验的基础上运用响应面分析的方法,对9种因素进行Plackett-Burman设计筛选得到3个显著因子,并对其进行最陡爬坡试验和Box-Behnken试验,利用Design-Expert V8.06分析软件进行回归分析得到最优组合。结果 优化后化学合成培养基的组成为:葡萄糖12g/L,麦芽糖18g/L,苏氨酸1.98g/L,50%乳酸钠0.5mL/L,K2HPO4·3H2O 0.3g/L,MgSO4·7H2O 0.5g/L,NaCl 0.5g/L,FeSO4·7H2O 0.01g/L,MnSO4·H2O 0.015g/L,MOPS 5g/L。结论 经验证,该培养基的产素能力比目前的清亮培养基提高了397.4%,并具有透明清亮且产素稳定等特点,为以后阿维链霉菌的理论和生产研究奠定了基础。 相似文献
