菝葜乙酸乙酯提取物抗癌活性的实验研究 总被引:2,自引:0,他引:2
目的 菝葜属百合科植物,为一种常用中药,目前所用的菝葜提取物多为其40%的乙醇提取物,本实验室首创了菝葜的乙酸乙酯提取工艺并对其进行了抗癌活性研究,旨在探讨其抗肿瘤作用及机制.方法 小鼠体内抗肿瘤试验和MTT法检测菝葜乙酸乙酯提取物在体内外对肿瘤细胞生长的抑制.结果 菝葜乙酸乙酯提取物可以在体内外有效抑制肿瘤细胞的增殖,对小鼠S180、H22、EAC肿瘤细胞及HepG2细胞株的半数抑制浓度(IC50)分别为(0.521±0.272)g/L、(0.801±0.333)g/L、(0.512±0.217)g/L、(0.608±0.268)g/L.结论 菝葜乙酸乙酯提取物具有抗肿瘤作用,具有临床应用的价值. 相似文献

小叶锦鸡儿中异黄酮及其苷类化合物的体外抗肿瘤活性研究 总被引:1,自引:0,他引:1
目的研究小叶锦鸡儿中异黄酮及其苷类化合物的体外抗肿瘤细胞增殖作用。方法利用四唑盐比色法检测,观察小叶锦鸡儿中分离出的3种异黄酮及其苷类化合物:雁靛黄素(Ⅰ)、高丽槐树-7-O-β-D-吡喃葡萄糖苷(Ⅱ)和芒柄花素-7-O-β-D-吡喃葡萄糖苷(Ⅲ)对人乳腺癌细胞株、宫颈癌细胞株、人肝癌细胞株生长的影响。结果化合物Ⅰ对人乳腺癌细胞株、宫颈癌细胞株的增殖均有明显的抑制作用,IC50分别为20.2、16.9μg.mL-1;化合物Ⅱ对乳腺癌细胞株、肝癌细胞株均有不同强度的抑制作用,IC50分别为17.9、18.8μg.mL-1;化合物Ⅲ对乳腺癌细胞株的增殖有抑制作用,但不呈现浓度依赖关系,对其他两个细胞株的增殖不显示抑制活性。结论化合物Ⅰ、Ⅱ对两种细胞株有不同程度的抗肿瘤活性,化合物Ⅲ不具抗肿瘤活性。 相似文献

采用复方金刚藤(金刚藤,大红藤,两面针,白花蛇舌草)治疗盆腔慢性炎症性包块30例,有效率达97%,与西药组33例对照,经Ridit检验,t=2.943,P〈0.1,2组疗效比较有显著性差异。复方金刚藤治疗效果明显优于西药,且无毒副作用,价格低廉,便于推广应用。 相似文献

目的:研究百合科菝葜属植物黑刺菝葜(Smilax scobinicaulis C.H.Wright)根脂溶性化学成分。方法:采用硅胶柱色谱、HP-20和Sephadex LH-20等分离手段对黑刺菝葜根脂溶性化学成分进行分离纯化,通过波谱数据分析(1H-NMR、13C-NMR)进行结构鉴定。结果:从脂溶性化学成分分离7个化合物。分别鉴定为:(1)β-sitosterol,(2)2,4-dihydroxy-3,6-dimethylbenzoate,(3)1-O-p-coumaroylglycerol,(4)ursolic acid,(5)blumenol A,(6)clemaphenolA,(7)1-O-feruloylglycerol。结论:化合物2~7为首次从该植物中分离得到。 相似文献

目的研究豆腐柴中总黄酮的最佳提取工艺及其抗菌作用效果。方法以提取时间、料液比、乙醇浓度分数作为自变量,以豆腐柴中总黄酮(按芦丁计)的提取率作为因变量,采用响应面法优选总黄酮的最佳提取工艺,并采用紫外分光光度法测定豆腐柴中总黄酮的含量。结果确定最优提取工艺为提取时间为25.95 min,料液比为1∶24.05,乙醇浓度为81.89%,提取次数为3次,在此条件下,豆腐柴中总黄酮得率理论上可达0.807%。结论星点设计-效应响应面法优选豆腐柴中总黄酮类的提取工艺,方法简单,结果可靠。 相似文献

Vascular dysfunction plays a key role in the pathogenesis of diabetic vascular disease. In this study, we aimed to investigate whether chronic in vivo treatment of Crataegus microphylla (CM) extract in diabetic rats induced with streptozotocin (STZ, intraperitoneal, 65 mg/kg) preserves vascular function and to evaluate whether the reduction of inducible nitric oxide synthase (iNOS), proinflammatory cytokines, and lipid peroxidation mediates its mechanisms of action. Starting at 4 weeks of diabetes, CM extract (100 mg/kg) was administrated to diabetic rats for 4 weeks. In aortic rings, relaxation to acetylcholine and vasoreactivity to noradrenaline were impaired, whereas aortic iNOS expression and plasma tumor necrosis factor‐α (TNF‐α) and interleukin‐6 (IL‐6), total nitrite–nitrate, and malondialdehite levels were increased in diabetic rats compared with controls. Chronic CM treatment significantly corrected all the above abnormalities in diabetic rats. In comparison, pretreatment of the aorta of diabetic rats with N‐[3(aminomethyl) benzyl]‐acetamidine, dihydrochloride (10–5 M), a selective inhibitor of iNOS, produced a similar recovery in vascular reactivity. These results suggest that chronic in vivo treatment of CM preserves endothelium‐dependent relaxation and vascular contraction in STZ‐induced diabetes, possibly by reducing iNOS expression in the aorta and by decreasing plasma levels of TNF‐α and IL‐6 and by preventing lipid peroxidation. Objective To investigate the effects of total alkaloids in Buxus microphylla leaves(ABML)on isolated rats thoracic aorta rings,and then to explore the possible mechanisms underlying the effects.Methods Thoracic aortas of Wistar rats were isolated,removed,and mounted onto an organ bath.The effects of ABML at different concentration on the contraction of isolated thoracic aorta rings(with and without endothelium)precontracted with KCl or PE were observed with organ bath technique.Dose-effect curves of CaCl2 were recorded by organ bath technique.The concentration of intracellular Ca 2+ ([Ca 2+ ]i)increased by PE,KCI,and caffeine in the presence of ABML was determined using Ca 2+ sensitive fluorescence indicator Fura-2/AM loaded thoracic aorta vascular smooth muscle (VSM)cells of rats.Results In aorta rings precontracted with PE and KCl,ABML produced concentration- dependent relaxation in both intact and denuded endothelium ring groups.There was no difference in the inhibition of contraction between the intact and denuded endothelium ring groups at the same concentration.Exposure of isolated thoracic aorta rings to ABML led to a significant reduction in the contracting response induced by CaCl2,and shifted the cumulative concentration-response curves to right.ABML could significantly inhibit the extracellular Ca 2+ influx induced by PE and KCl under[Ca 2+ ]0 of 1.5 mmol/L,with inhibitory ratios of 40.2%and 49.9%,respectively.In the case of Ca 2+ -free,ABML could significantly inhibit the intracellular Ca 2+ release induced by PE,with inhibitory ratio of 72.4%.Conclusion ABML relaxes thoracic aorta VSM cells by suppressing influx of extracellular Ca 2+ via voltage-dependent Ca 2+ channel and receptor-operated Ca 2+ channel. 相似文献
