Glutamate-induced currents were recorded from cultured trout astrocytes with the whole-cell variation of the patch-clamp technique. Ninety percent of the tested cells were directly depolarized by the amino acid neurotransmitter in a concentration-dependent manner. The depolarizing effect was due to an inward current that reversed near 0 mV and was accompanied by a noise increase, indicating the opening of an ion channel. Ion substitution experiments revealed that the glutamate-induced current was mainly carried by sodium ions but not chloride or calcium ions. The glutamate-induced response could be mimicked by the neuronal glutamate receptor subtype agonists kainate and quisqualate, while N-methyl-D-aspartate was without detectable effect. © 1995 Wiley-Liss, Inc.  相似文献

During June and July of 2007, a total of 130 specimens of the fish Rastrelliger kanagurta Cuvier (Teleostei, Scombridae), ranging between 19–31 cm in total length, were caught in the Red Sea off the coast of Sharm El-Sheikh, South Sinai, Egypt, and examined for infections by acanthocephalans (65 fish/month). Of this number, 29 (22.30%) were slightly or heavily parasitized by the acanthocephalan Diplosentis nudus (Harada, 1938) Pichelin et Cribb, 2001 (Cavisomidae); no other helminth parasites were found in the intestine of R. kanagurta. Twenty-nine infrapopulations of D. nudus, ranging from 23–218 individuals were collected from the infected fishes. These infrapopulations were distributed in a well-defined fundamental niche along the intestine of R. kanagurta, where the distribution of male worms was not random with respect to female worms size and position and suggests that the male-male competition for access to female may be intense and may select for large males. No correlation between fish size and infrapopulation size was observed. Correlations between female-to-male sex ratio and infrapopulation size, numbers of females and their mean lengths, numbers of males and their mean lengths, mean female length and mean male length within infrapopulation were very strong, and clearly suggest that as the infrapopulation size increased, the number of females and their mean lengths decreased and the number of males and their mean lengths increased. Combination of these results strongly suggests density-dependent effects and competition between male worms. The relationship between the mean female length or size and the number of eggs within its pseudocoel was strongly positive; egg production by female worm significantly decreases as the infrapopulation size increases, suggesting density-dependent reduction in female worm fecundity. Tendency for the variability in male testes size was not significant in infrapopulations of D. nudus. All of these results are discussed.  相似文献

In fishes, exploitation of aerial gas exchange has evolved independently many times, involving a variety of air-breathing organs. Indeed, air-breathing occurs in at least 49 known families of fish (Graham, 1997). Many amphibious vertebrates, at some stage of their development are actually trimodal breathers that use various combinations of respiratory surfaces to breath both water (skin and/or gill) and air (skin and/or lung). The present review examines the evolutionary implications of air-breathing organs in fishes and the morphology of the peripheral receptors and the neurotransmitter content of the cells involved in the control of air-breathing. Control of breathing, whether gill ventilation or air-breathing, is influenced by feedback from peripheral and/or central nervous system receptors that respond to changes in PO2, PCO2 and/or pH. Although the specific chemoreceptors mediating the respiratory reflexes have not been conclusively identified, studies in water-breathing teleosts have implicated the neuroepithelial cells (NECs) existing in gill tissues as the O2 sensitive chemoreceptors that initiate the cardiorespiratory reflexes in aquatic vertebrates. Some of the air-breathing fishes, such as Protopterus, Polypterus and Amia have been shown to have NECs in the gills and/or lungs, although the role of these receptors and their innervation in the control of breathing is not known. NECs have been also reported in the specialized respiratory epithelia of accessory respiratory organs (ARO’s) of some catfish species and in the gill and skin of the mudskipper Periophthalmodon schlosseri. Unlike teleosts matching an O2-oriented ventilation to ambient O2 levels, lungfishes have central and peripheral H+/CO2 receptors that control the acid-base status of the blood.  相似文献

目的 了解河南省淡水养殖环节中非O1/O139群霍乱弧菌毒力基因分布及分子分型情况。方法 对河南省淡水养殖环节中50株非O1/O139群霍乱弧菌和3株病人来源菌株进行全基因测序,利用PubMLST-Vc数据库分析其序列分型(ST),利用最小生成树关系图分析进化关系,通过VFDB数据库获得其毒力基因分布。结果 来源于淡水养殖环节和来源于病人的非O1/O139群霍乱弧菌53株菌株均具有黏附、趋化运动、抗吞噬、毒素及酶类等功能的8类毒力相关因子基因,缺失辅助定植、毒素共调、分泌等功能的毒力相关因子基因和副霍乱肠毒素等4个毒素基因。部分毒力相关因子或部分菌株的毒力相关因子毒力基因不全。与3株来源于病人的菌株相比,二者毒力因子基因携带情况相近,除MSHA Ⅳ型菌毛毒力因子mshA基因、荚膜多糖wbuB基因、wbfY基因、rmlB基因、血红素受体hutA基因及Ⅲ型分泌系统vscC2和vcrD2基因外,部分菌株二者携带相同的毒力因子基因。53株非O1/O139群霍乱弧菌分属19个ST型,ST4和ST5是优势ST型,养殖环节来源的菌株与病人来源的菌株分属不同的ST型。19个ST型等位基因位点变异差异数在1~7个,其中分离自淡水养殖环节的非O1/O139群霍乱弧菌菌株17个ST型等位基因位点变异差异数在1~7个,病人来源的非O1/O139群霍乱弧菌菌株2个ST型与分离自淡水养殖环节的非O1/O139群霍乱弧菌菌株ST1、ST2、ST6和ST10属同一簇,与ST1有6个等位基因位点存在差异。结论 河南省淡水养殖环节非O1/O139群霍乱弧菌菌株MLST分型多样化,携带多种毒力相关因子,不同来源菌株携带的毒力基因相同,虽然分属不同的ST型,但还是存在食品安全风险,提醒有关部门采取措施进行防控。  相似文献

目的 了解海南省淡水鱼中食源性寄生虫感染情况,评估人群感染潜在的危险性,为制定食源性寄生虫病监测策略和防治方案提供科学依据。方法 2019—2021年对海南省琼中县、三亚市和昌江县监测点附近的溪流或水库等自然水体环境,以随机捕捞的方式获取新鲜样品。采用玻片压片镜检法对寄生虫囊蚴进行检测。同时在捕捞样品的行政村进行肝吸虫病知识和居民肝吸虫病感染情况调查。结果 2019—2021年共检测淡水鱼47种合计396尾,检出寄生虫囊蚴阳性45尾,总感染率为11.36%。有13种淡水鱼中检出寄生虫,品种感染率为27.66%(13/47)。其中海南细齿塘鳢感染率最高,为75.00%(3/4),显著高于其他鱼种(P<0.05),三个市县中,琼中县检出率最高,为17.05%(22/129)。发放调查问卷487份,居民对肝吸虫病总知晓率为11.50%(56/487),昌江县知晓率最低,为1.31%,男性知晓率高于女性(P<0.05)。同期对居民肠道进行寄生虫检测3 178人份,未发现肝吸虫病患者。结论 海南省部分市县淡水鱼受到寄生虫污染,应加强监测,采取有效的防治措施,加强食源性寄生虫病卫生宣传和教育,提高群众自我防护意识。  相似文献

Survival of juvenile freshwater mussels (Echyridella menziesii (Gray, 1843) formerly known as Hyridella menziesi) and crayfish (Paranephrops planifrons, White, 1842) decreased after four days exposure to microcystin‐containing cell‐free extracts (MCFE) of Microcystis sp. at concentrations typical of severe cyanobacterial blooms. Crayfish survival was 100, 80, and 50% in microcystin concentrations of 1339, 2426, and 11146 μg L?1 respectively, and shade‐ and shelter‐seeking behavior was negatively affected when concentrations were ≥2426 μg L?1. Mussel survival decreased to 92% and reburial rates decreased to 16% after exposure for 96 h to MCFE containing microcystins at concentrations of 5300 μg L?1. Crayfish survival was 100% when fed freeze‐dried Microcystis sp. incorporated into an artificial diet (6–100 μg microcystin kg?1 ww) at dietary doses from 0.03 to 0.55 μg g?1 body weight d?1 for 27 days. Specific growth rate was significantly lower in crayfish fed ≥0.15 μg g?1 body weight day?1 compared with controls, but not compared with a diet incorporating nontoxic cyanobacteria. Microcystins accumulated preferentially in crayfish hepatopancreas and mussel digesta as MCFE or dietary concentrations increased. These laboratory data indicate that, assuming dissolved oxygen concentrations remain adequate, and no simultaneous exposure to live Microcystis sp. cells, cell‐free microcystins will only be a significant stressor to juvenile crayfish and mussels in severe Microcystis sp. blooms. In contrast, crayfish were negatively affected by relatively low concentrations of microcystins in artificial diets compared with those measured locally in benthic cyanobacterial mats. © 2012 Wiley Periodicals, Inc. Many teleost fishes with no apparent modifications for life on land are able to produce effective terrestrial locomotor behaviors, including a ballistic behavior called the “tail-flip” jump. Cyprinodontiformes (killifishes, Teleostei: Atherinomorpha) that live at the water's edge vary in morphology and inclination to emerge onto land. Do fish with an amphibious predisposition have extensive modification of the propulsive region of the body when compared to fully aquatic relatives? We quantified body shape and anatomy of the caudal peduncle and tail (the propulsive organ on land and in water) in 11 cyprinodontiform species and two outgroup taxa (Atherinomorpha). We hypothesized that amphibious species would have longer, “shallower” bodies (larger body fineness ratios), deeper (proportionally larger) caudal peduncles, and more robust bones in the tail fin (larger ossified area of the hypural/epural bones) to facilitate locomotor movements on land. We found no evidence of convergence in body shape or skeletal anatomy among species known to make voluntary sojourns onto land. In fact, deep-bodied species, shallow-bodied species, and species with intermediate morphologies all are able to emerge from the water and move on land. It is possible that there are as-yet-undocumented subtle soft-tissue (muscle, tendon, and ligament) modifications that enhance terrestrial locomotor performance in species known to spend large periods of time on land. However, it is also possible that extreme anatomical changes are not required for aquatic cyprinodontiform species to produce effective locomotor movements when they emerge out of the water and move across the land. Cyanobacterial blooms are a worldwide ecological issue. Cyanophages are aquatic viruses specifically infecting cyanobacteria. Little is known about freshwater cyanophages. In this study, a freshwater cyanophage, Mae-Yong924-1, was isolated by the double-layer agar plate method using Microcystis aeruginosa FACHB-924 as an indicator host. Mae-Yong924-1 has several unusual characteristics: a unique shape, cross-taxonomic order infectivity and a very unique genome sequence. Mae-Yong924-1 contains a nearly spherical head of about 100 nm in diameter. The tail or tail-like structure (approximately 40 nm in length) is like the tassel of a round Chinese lantern. It could lyse six diverse cyanobacteria strains across three orders including Chroococcales, Nostocales and Oscillatoriales. The genome of the cyanophage is 40,325 bp in length, with a G + C content of 48.32%, and 59 predicted open reading frames (ORFs), only 12 (20%) of which were functionally annotated. Both BLASTn and BLASTx scanning resulted in “No significant similarity found”, i.e., the Mae-Yong924-1 genome shared extremely low homology with sequences in NCBI databases. Mae-Yong924-1 formed a root node alone and monopolized a root branch in the proteomic tree based on genome-wide sequence similarities. The results suggest that Mae-Yong924-1 may reveal a new unknown family apparently distinct from other viruses.  相似文献

深圳市淡水鱼华支睾吸虫感染调查及实验研究   总被引:7,自引:5,他引:7  
目的了解深圳市淡水鱼华支睾吸虫(肝吸虫)囊蚴感染情况,阻断和控制肝吸虫病在深圳市的传播和流行。方法用鱼体消化法筛查肝吸虫囊蚴;以肝吸虫过氧化物酶(SOD)基因保守序列作为摸板,RT- PCR鉴别肝吸虫囊蚴。结果对深圳市淡水鱼肝吸虫感染情况进行调查,共抽检16种鱼257份样品,其中检出肝吸虫囊蚴45份,检出率为17.50%。检出率最高的鱼种依次为鲩鱼40.74%、鲮鱼26%、鲤鱼22.5%、大头鱼 22.85%、生鱼16.6%、非洲鲫鱼14.5%;RT-PCR检测结果与镜检结果完全一致。结论检测结果显示深圳市淡水鱼中肝吸虫的感染情况严重,尤其是鲩鱼的感染率较高,为了控制和阻断肝吸虫的传播必须改变吃淡水鱼生的生活习惯。  相似文献
