首先对人和小鼠的TCRVβ的氨基酸序列进行多序列对准,就Vβ之HV4片段进行比较,分析与超抗原TSST-1结合的四种Vβ(小鼠Vβ3、Vβ15、Vβ17和人Vβ2)之HV4序列内是否存在特定的氨基酸残基排列主型。结果发现:小鼠Vβ3和Vβ17的HV4具有特异的RFSAXCXSNS主型,而小鼠Vβ15和人β2的HV4则含独特的KFXIXH主型。提示:与TSST-1结合的四种Vβ所对应的T细胞识别表位可能不止一个。  相似文献

目的:探讨艾滋病免疫重建不全患者的TCRVβ基因多样性改变及中药免疫2号方的干预作用.方法:采集37例艾滋病免疫重建不全患者治疗前后的外周血PBMC,另以15例HIV抗体阴性健康献血员做对照,采用人类TCRVβ基因CDR3多样性定量检测试剂盒检测,基因扫描作图并计算Vβ家族中每个家族不同大小CDR3区片段的分布.结果:HAART治疗后1年免疫重建不全患者的TCRVβ多个家族的扫描图高斯分布被打破,并发生TCR谱系的偏移,经半年中药联合HAART治疗后,偏移的TCR谱系有所恢复.以试剂盒提供的定量分析软件计算的距离D(distance)值来量化评价,2组D值改变无明显差异,但联合中药能减少数据的变异性.CD4+T细胞计数与TCRVβ基因多样性改变两者存在显著的相关关系(r=-0.772,P=0.000).结论:中药复方对于TCRVβ各家族单寡克隆情况有不同程度的改善和恢复,提示中医药可能促进T细胞部分受体基因重排,丰富受体库,帮助机体免疫细胞有效识别病毒,减少T细胞凋亡.  相似文献

A specialized and fine-tuned immune response of bats upon infection with viruses is believed to provide the basis for a “friendly” coexistence with these pathogens, which are often lethal for humans and other mammals. First insights into the immunity of bats suggest that bats have evolved to possess their own strategies to cope with viral infections. Yet, the molecular details for this innocuous coexistence remain poorly described and bat infection models are the key to unveiling these secrets. In Jamaican fruit bats (Artibeus jamaicensis), a New World bat species, infection experiments with its (putative) natural viral pathogens Tacaribe virus (TCRV), rabies virus (RABV), and the bat influenza A virus (IAV) H18N11, have contributed to an accurate, though still incomplete, representation of the bat-imposed immunity. Surprisingly, though many aspects of their innate and adaptive immune responses differ from that of the human immune response, such as a contraction of the IFN locus and reduction in the number of immunoglobulin subclasses, variations could also be observed between Jamaican fruit bats and other bat species.  相似文献

Analysis of TCR beta-chain complementarity-determining region size gives an indication of the T cell immune response. We examined CD4+ and CD8+ subgroups of T cells in the peripheral blood (PBL), benign gastric mucosa, and tumor (TIL) lymphocytes of 12 patients with primary gastric carcinomas of both intestinal and diffuse types. The average number of expanded clones in each compartment, expressed by the 24 families of the TCRVbeta repertoire, was analyzed according to tumor histological type, maximal invasive depth, and lymph node metastases. Fewer clones were expressed by the PBL in the cases with lymph node metastases than in those without (CD4+ P = 0.00017, CD8+ P = 0.016). Fewer CD8+ clones were expressed by the PBL in the cases with full thickness tumor infiltration than in those involving only the mucosa and submucosa (P = 0.05). The CD8+ TIL showed significantly fewer clones in the diffuse-type carcinoma than in the intestinal type (P = 0.046).  相似文献

目的 探讨γδT细胞在哮喘气道慢性炎症产生或延续机制中的作用。方法 留取 7例哮喘患者和 7例对照者的外周血和支气管肺泡灌洗液 (BALF)。用免疫荧光染色和流式细胞分析技术检测γδT细胞的百分率 ,用RT PCR法和基因扫描法分析T细胞受体Vδ1~Vδ3 亚族的相对取用频率及克隆增殖状态。结果 哮喘患者BALF中γδT细胞比例 (7 8%± 4 7%)较对照组 (3 3%± 3 0 %)显著增加 (P =0 0 4 )。哮喘患者的外周血及BALF中V1~Vδ3 亚族的相对取用频率无显著差异 ,但BALF中Vδ1的相对取用频率 (44%± 13%)较对照组 (19%± 5 %)显著增加 (P =0 0 0 0 2 )。哮喘患者BALF中部分Vδ基因亚家族呈单克隆或寡克隆扩增 ,以Vδ1亚族尤其显著。结论 对单一或数种抗原发生特异性应答的γδT细胞在哮喘的炎症过程中可能发挥了较重要的作用 ,而这种 /些抗原的持续刺激可能是支气管哮喘气道炎症发生与持续发展的原因之一。  相似文献
