目的:检测神经纤维瘤蛋白在先天性脊柱侧凸患者成骨细胞和软骨细胞中的表达。方法:6例先天性脊柱侧凸患者,在后路手术时取髂骨及髂骨生长板,分离、培养成骨细胞和软骨细胞,分别行碱性磷酸酶染色和甲苯胺蓝染色。逆转录-多聚酶链反应(RT—PCR)检测神经纤维瘤蛋白mRNA.间接免疫荧光和Westemblot检测神经纤维瘤蛋白在成骨细胞和软骨细胞中的表达。结果:先天性脊柱侧凸患者成骨细胞和软骨细胞中存在Ⅱ型神经纤维瘤蛋白表达,该蛋白主要分布在细胞浆,所表达蛋白为三磷酸鸟苷酶活化蛋白(GAP)活性较弱的Ⅱ型异构体。结论:先天性脊柱侧凸患者成骨细胞和软骨细胞中存在神经纤维瘤蛋白表达,但该蛋白是否通过对成骨细胞和软骨细胞的影响导致骨骼系统异常还有待于进一步研究。 相似文献

PTH has diverse effects on bone metabolism: anabolic when given intermittently, catabolic when given continuously. The cellular
mechanisms underlying the varying target cell response are not clear yet. PTH induces RGS-2, a member of the Regulator of
G-protein Signaling protein family, via cAMP/PKA, and inactivates PKC-mediated signaling. To investigate intracellular signaling
pathways with different PTH concentration-time patterns, we treated UMR 106-01 osteoblast-like cells in a perfusion system.
PTH was administered intermittently (4 min/h, 10−7 M) or continuously at an equivalent cumulative dose (6.6 × 10−9 M). cAMP was measured using radioimmunoassay, mRNA levels using real-time rtPCR and ribonuclease protection assay, and protein
levels using Western immunoblotting. A single PTH pulse transiently increased cAMP levels by 2000% ± 1200%. In contrast to
continuous PTH exposure, cAMP induction remained unchanged with intermittent PTH, ruling out desensitization of the PTH receptor.
In continuously perfused cells, RGS-2 abundance was three to five times higher than in cells intermittently exposed to PTH
for up to 12 h. MKP-1 and -3 were significantly less induced with pulsatile PTH; exposure-mode-dependent differences in MMP-13
and IGFBP-5 were small. Pulsatile but not continuous PTH administration prevents PTHrP receptor desensitization and accumulation
of RGS-2 in osteoblasts, which should preserve PKC-dependent signaling. 相似文献

Summary Two methods for harvesting osteoblast-like cell populations from newborn (10 days) rat calvaria were compared. The first one
consisted in culturing the periosteum-free bones and then trypsinizing the cells on the bone surface. The second one involved
the migration of the osteoblasts on glass fragments before trypsinization. Since the plating efficiency, the proportion of
alkaline phosphatase-positive cells, the population doubling time, and the calcium deposition were more adequate, the second
method was used to further characterize the behavior of the cultures. During the first week of culture, the cells featured
shapes similar to those observedin vivo on the surface of periosteum-free calvaria. They formed multilayers and, in the presence of ascorbic acid, synthetized an
organic matrix containing exclusively type I collagen. Later, small amounts of type III collagen appeared. The cells were
embedded in the matrix and progressively acquired the morphologic phenotype of osteocyte-like cells. The matrix mineralized
in the presence of β-glycerophosphate. The technique of dropinoculation (high concentration of cells in a small volume of

Implant surface morphology regulates osteoblast phenotypic expression. Osteoblast sensitivity to non-biologic surfaces suggests that native bone surface features may also affect osteoblast response. To test this, MG63 osteoblast-like cells were grown for 7 days on bovine cortical bone wafers pretreated with rat bone marrow osteoclasts for 0, 10 or 20 days. Response to osteoclast-treated surfaces was compared to the response of MG63 cells to titanium surfaces with smooth and rough microtopographies. Cell number, differentiation (alkaline phosphatase activity and osteocalcin levels), and local factors (PGE(2) and TGF-beta1) were measured in confluent cultures. Compared to culture on plastic, cell number was reduced on all three types of bone wafers; this effect was dose-dependent with increasing resorption of the surface. 探讨周期性双轴力学应变对成骨细胞增殖与分化合成功能的影响。将正常 3月龄雌性 SD大鼠和骨质疏松大鼠颅顶骨分离的成骨细胞分别在含 10 %胎牛血清的 F- 12培养液中培养 ,并接种在双轴力学应变装置中。当细胞生长至亚融合状态 (Subconfluence) ,给细胞施加力学刺激 ,频率为 1Hz,力学刺激分别为 4 0 0、10 0 0、4 0 0 0μ strain;作用时间分别为每天 30 m in,2、4、8h,共加载两天。以未受力学刺激的细胞为对照组 ,受力学刺激的细胞为实验组 ,并进行比较。采用流式细胞技术测定细胞增殖变化 ;采用同位素标记方法检测成骨细胞骨钙素、I型胶原 C端前肽 (PICP)和总蛋白的分泌量。结果表明 :1)在静态培养条件下 ,3ovx组与 3control组比较 ,其细胞功能活性无明显变化 ,但 3ovx组大鼠成骨细胞增殖活性明显增高 ,这与绝经后骨质疏松骨代谢的高转换率相一致。 2 )4 0 0、10 0 0 μstrain力学刺激可以促进 3control组成骨细胞 I型胶原、骨钙素和总蛋白的分泌量增加 ,促进成骨细胞的分化成熟 ;在 10 0 0μstrain力学刺激下 ,成骨细胞合成骨基质的能力增加最为明显。同时 ,在 4 0 0、10 0 0μstrain力学刺激的初期也可以促进成骨细胞的增殖 ,而促进成骨细胞的分化成熟的作用大于促进细胞增殖的作用。 3)在4 0 0 0 μ 相似文献

目的 对比葛根素和淫羊藿甙对成骨细胞增殖和分化的影响。方法 将小鼠骨髓基质干细胞定向诱导分化为成骨细胞,取第3、4代细胞,以rhBMP-2为阳性对照,MTT法观察葛根素和淫羊藿甙对成骨细胞的增殖作用,碱性磷酸酶(ALP)比活性测定及钙结节计数观察葛根素和淫羊藿甙对成骨细胞分化的影响。结果 20μg/ml的淫羊藿甙可显著促进成骨细胞的增殖;20μg/ml淫羊藿甙及50μg/ml葛根素均能提高成骨细胞的碱性磷酸酶活性,并使矿化结节的形成明显增多。结论 淫羊藿甙不仅能促进成骨细胞的分化,还能增强其增殖能力,而葛根素主要是通过刺激分化来增强成骨细胞的活力。 相似文献

目的:在H2O2胁迫下,研究虾青素增强成骨细胞抗自由基损伤的作用。方法:用H2O2作用MC3T3-E1成骨细胞,建立自由基损伤细胞模型。培养的成骨细胞分为对照组、模型组、虾青素低剂量组(1×10^-7mol/L)、虾青素中剂量组(1×10^-6mol/L)和虾青素高剂量组(1×10^-5mol/L)。测定不同处理组细胞活力、超氧化物歧化酶(superoxide dismutase,SOD)活性、活性自由氧(reactive oxygen species,ROS)含量、脂质过氧化物(lipid oxygen,IJP0)含量,同时利用荧光偏振法测定细胞膜流动性。结果:模型组细胞活力、SOD活性、细胞膜流动性均比各剂量虾青素组显著降低(P〈0.01),而ROS、LPO含量均比各剂量虾青素组显著升高(P〈0.01)。结论:H2O2摄入会导致大鼠成骨细胞的氧化损伤,而虾青素可以预防或降低此类损伤对细胞的影响。 相似文献

Objective: To elucidate the effects of exogenous basic fibroblast growth factor ( bFGF ) on biological characteristics of rat osteoblasts cultured in vitro. Methods: The osteoblasts isolated from a Sprague-Dawley rat and cultured in vitro were treated with different concentrations of bFGF ( 5-50 ng/ml) respectively. At 24 hours after treatment, the proliferating cell nuclear antigen was measured with immunocytochemistry, alkaline phosphatase (ALP) activity was determined and the expression of transforming growth factor beta 1 ( TGF-β1 )was detected to observe the effects of bFGF on growth and differentiation of osteoblasts. Resu/ts: bFGF ( 5-50 ng/ml ) could obviously promote the growth of osteoblasts. The intracellular expression of TGF-β1 mRNA increased significantly, but the intracellular ALP content decreased. Conclusions: bFGF can obviously stimulate the proliferation of osteoblasts and promote the synthesis of TGF-β1, but cannot promote the differentiation of osteoblasts. 相似文献

目的探讨成骨细胞与血管内皮细胞联合培养的生物学特性. 方法取2周龄乳兔颅盖骨及肾脏皮质传代培养制备成骨细胞(A组)、血管内皮细胞(B组)及成骨细胞与血管内皮细胞联合培养(C组),用Ⅰ型胶原和血管Ⅷ因子免疫细胞化学染色鉴定成骨细胞和血管内皮细胞,倒置相差显微镜和组织学染色观察细胞的生长特性和细胞相容性,检测碱性磷酸酶 (alkaline phosphatase,ALP)活性,观察血管内皮细胞对成骨细胞产生的ALP活性有无影响,MTT法检测细胞活力,分析细胞生长和增殖情况. 结果免疫细胞化学染色证实,培养的细胞为成骨细胞和血管内皮细胞.倒置相差显微镜、HE和Masson染色均显示两种细胞混合生长良好.ALP检测结果:C组ALP活性明显高于A组和B组(P<0.01),A组高于B组(P<0.05).MTT检测结果表明:C组细胞早期增殖较慢,而后期增殖较快. 结论成骨细胞与血管内皮细胞具有良好的相容性,血管内皮细胞能够增强成骨细胞的ALP活性,提高成骨细胞的增殖能力.联合培养细胞具有很强的增殖潜能. 相似文献
